AQP1在非小细胞肺癌中的表达及其与MMP-9、Ki67表达的关系

【摘要】 目的探讨水通道蛋白-1(AQP1)在非小细胞肺癌(NSCLC)中的表达和意义,以及与基质金属蛋白酶-9(MMP-9)、细胞核增殖相关抗原(Ki67)表达的关系。方法采用免疫组织化学方法检测50例非小细胞肺癌组织中AQP1和MMP-9、Ki67的表达,同时随机选择30例癌旁正常肺组织、30例肺良性肿物作对照。分析AQP1表达与临床病理特征以及与MMP-9、Ki67表达的相关性。结果1.AQP1阳性物质定位于胞浆和/或胞膜,亦见细胞核着色,瘤体边缘与正常组织交界处表达增多;AQP1蛋白在NSCLC、癌旁正常肺组织、良性肿物中的阳性率分别为32.0%(16/50)、3.3%(1/30)和6.6%(2/30),差异有显著统计学意义(P<0.01)。2.AQP1蛋白在NSCLC中的表达与性别、年龄、临床分期、肿瘤T分期无关(P>O.05),与病理类型、分化程度、是否有淋巴结转移及是否有复发转移有关(P<O.05)。3.AQP1和MMP-9在NSCLC中的表达有相关性(P<O.05,P=O.030);AQP1与Ki67在NSCLC中的表达亦相关(P<O.05,P=O.011)。结论1.在NSCLC中,AQP1的表达水平明显高于癌旁正常肺组织,但与肺良性肿物间无差异。2.AQP1在NSCLC中的表达与病理类型、分化程度和是否淋巴结转移及是否复发转移密切相关,而与性别、年龄、临床分期、T分期无关,提示AQP1在NSCLC的侵袭转移中可能具有重要的作用。3.AQP1和MMP-9的表达正相关,提示AQP1和MMP-9在NSCLC中的侵袭、转移过程中可能有相互促进的作用;AQP1和Ki67的表达正相关,提示AQP1和Ki67在NSCLC中的增殖过程中可能有相互促进的作用。4.联合检测AQP1、MMP-9及Ki67可作为判定非小细胞肺癌恶性程度,评估其侵袭性及转移性的重要指标。5.以AQP1作为干预靶点来抑制癌细胞的增殖、浸润及转移,可能是治疗NSCLC的新途径。

【Abstract】 ob j eetive DISeuSSion of the water ehannel protein-1(AQPI)innon-Small eell lung eaneer(NSCLC)in the expreSSion and Signifieanee,as well as matrix metalloproteinase-9(MMP-9),proliferating eellnuelear antigen(Ki67)expreSSion.Methods Deteeted by inununohistoehemistry 50 eases of non-smalleell lung eaneer AQPI and MMP-9,Ki67 exPression,and randomlyseleeted 30 eases of adjaeent normal lung tissue,30 eases ofPulmonary benign masses as a eontrol.AQPI exPression analysis ofelinieal and Pathologieal features as well as MMP-9,Ki67 exPressioneorrelation.Results 1.AQPI Positive substanee loeated in the eytoPlasm and/or membrane,nueleus eoloring also see the tumor and normal tissueat the junetion of the edge inereased exPression:AQPI Protein inNSCLC,adjaeent normal lung tissue,benign tumor,the positive rate32.0%(16/50),3.3%(1/30)and 6.6%(2/30),the differenee wassignifieant(p<0.01). 2.AQPI Drotein exDression in NSCLC,sex,age, clinical stage, tumor T staging(P>O.05), and the pathological type,differentiation degree, lymph node metastasis and whether there arerecurrent metastasis(P<O.05). 3.AQP1 and MMP-9 expression in NSCLCwas correlated(P<O.05, P=O.030); AQP1 and Ki67 expression in NSCLC is also associated(P<O.05, P=O.011).Conclusions 1.AQP1 in NSCLC, significantly higher than cancer expression level of normal lung tissue, but by with no differencebetween benign neoplasm lung. 2.AQP1 expression in NSCLC withpathological type, differentiation, and whether the lymph node metastasis and recurrence and metastasis are closely related, butnot with sex, age, clinical stage, T staging, suggesting that AQP1 in NSCLC invasion and metastasis may have important role. 3.AQP1 andthe expression of MMP-9 positively correlated, suggesting that AQP1 and MMP-9 in NSCLC in the invasion and metastasis may be mutually reinforcing; AQP1 and the expression of Ki67 positive correlation,suggesting that AQP1 and Ki67 in the NSCLC proliferation may have mutually reinforcing effects. 4. Combined detection of AQP1, MMP-9 and Ki67 can be used to determine the degree of malignancy ofnon-small cell lung cancer, to assess an important indicator of invasive and metastatic. 5. AQP1 as the intervention target toinhibit cancer cell proliferation, invasion and metastasis, may be a new way to treat NSCLC.
