艾灸对实验性RA滑膜细胞GC信号通路影响的研究

【摘要】 目的:在确证艾灸对实验性RA家兔抗炎效应的基础上,观察艾灸对实验性RA家兔滑膜细胞糖皮质激素(GC)信号通路的影响,揭示艾灸治疗实验性RA抗炎效应的分子信号机制。方法:日本大耳白兔36只,按体重、性别分层随机分为空白对照组(简称空白组)、RA模型组(简称模型组)和艾灸治疗组(简称治疗组),每组12只动物。将福氏完全佐剂(FCA)按0.5ml/Kg体重剂量平均注入模型组、治疗组各动物双后膝关节腔内,空白组各动物采用相同方法注入等容积无菌生理盐水作为对照。艾粒灸双侧“肾俞”穴各五壮,每日一次,6天为一个疗程,共治疗3个疗程,疗程之间休息一天。空白组和模型组动物同法固定,但不进行艾灸治疗。用软皮尺于造模前及治疗后第3、6、9、12、15、18、21天分别测定各家兔左右后膝关节周长。治疗后第21天处死家兔,取后膝关节滑膜组织,运用基因芯片及生物信息分析技术检测、分析滑膜细胞糖皮质激素(GC)信号通路中36种相关信号分子的表达。结果:造模前,各组家兔左、右后膝关节周长无显著性差异(P>0.05);造模后各个时间点模型组左、右膝关节周长均较正常组明显升高(P<0.01),说明炎症模型持续存在:治疗后艾灸组左、右膝关节周长均较模型组下降(P<0.05),提示艾灸具有抗炎消肿作用。造模组与空白组比较,GC信号通路异常激活,大量促进炎症的相关信号分子表达上调;治疗组与造模组比较,GC通路部分抑制炎症的相关信号分子表达上调,部分信号分子虽无显著上调或下调(无明显统计学意义),但与抑炎相关的信号分子有上调趋势,与促炎相关的信号分子有下调趋势。结论:艾灸具有抗炎消肿作用,艾灸对实验性RA滑膜细胞糖皮质激素(GC)信号通路的调控可能是其发挥抗炎作用的重要机制。

【Abstract】 Objective:Observe the influence of GC pathway’s signal transduction in RA synovial cell of experimental rabbits by moxibustion,and reveal the anti-inflammatory molecular signal mechanism of experimental RA by moxibustion.Methods:Apply gene chip and bioinformation analytical technique to detect the 36 sorts of signal molecule of GC pathway.Results:Compare model group with normal group, GC pathway has been activated abnormally, and as a result, the expression of proinflammatory signal molecules has been up-regulated; compare moxibustion group with model group, the expression of some anti-inflammatory signal molecules has been up-regulated, while some signal molecules showed no significant increases or reduction(Results are not statistically significant), however, the expression of some anti-inflammatory signal molecules show an upward tendency, and the expression of some proinflammatory signal molecules show an downward tendency.Conclusions:Moxibustion plays an important roll in regulating GC pathway’s signal transduction in RA synovial cell of experimental rabbits. Maybe this is an important mechanism of Moxibustion playing anti-inflammatory effect.
