补肾填精法调控辐射小鼠骨髓ATM/ATR通路的研究

【摘要】 辐射对人类的危害越来越受到重视,辐射虽可导致人体免疫、血液、生殖及其他多系统损害,但其根本原因是导致DNA这一重要射线靶的损伤,本文从外感热病角度研究补肾填精法调控辐射损伤的作用机理。目的:观察补肾填精方对Co60γ射线所致辐射损伤小鼠模型的作用,并通过观察其对DNA损伤修复ATM/ATR通路的影响,探讨该方抗辐射的作用机制。方法:将雌雄各半的balb/c小鼠,分成正常组,病理对照组,补肾组,补血组及补阴组。采用Co60γ射线损伤小鼠模型,在造模后取骨髓运用免疫组化方法对关键基因ATM/ATR信号通路上相关蛋白表达结合图像分析技术进行分析;摘取动物眼球取外周血,对其外周血进行血常规分析;造模后取睾丸和卵巢,采用HE染色观察其病理形态学。结果:1.免疫组化结果:补肾组和补血组与病理组相比在ATR蛋白表达方面存在差异(P<0.05)而ATM基因表达在各组中的差异不明显。2.生殖系统HE染色结果:睾丸的补肾组受损程度与模型组比较,差别较大,修复更为明显;补血和补阴组受损程度与模型组比较较轻;卵巢的补肾组,补血组和补阴组大多数颗粒细胞外层保留,内层出现部分坏死、脱落,部分卵母细胞损伤,受损程度均较模型组轻。3.外周血结果:白细胞总数,中性粒细胞,补肾组和补血组与病理组有差异(P<0.05),淋巴细胞来看,补阴组和病理组有差异(P<0.05)而血红蛋白来看,3个组和病理组均有差异(P<0.05)。结论:1.补肾填精方可促进ATR基因的表达增强;2.补肾填精方在一定程度上对受照小鼠生殖功能具有保护作用;3.补肾填精方对于放射后小鼠骨髓细胞的修复都有一定的促进作用,其对促进白细胞增生较为明显。综上补肾填精方具有抗辐射,保护小鼠生殖功能,促进血细胞生长的作用。

【Abstract】 Radiation hazards to humans more and more attention, although radiation can cause immune, blood, reproductive and other multi-system damage, but the fundamental reason leading to this important radiation target DNA damage, From the outside are fever perspective study invigorating the kidney and fill in a law to regulate radiation damage to the role of the mechanism.Aims:Observed Bushentianjing side Co60y ray radiation injury in mice induced by the role model, and by looking at its DNA damage repair ATM/ATR pathway, anti-radiation of the party mechanism.Methods:The male and female half and half balb/c mice were divided into normal group, pathological control group, kidney group, blood group and yin group. Ray irradiated mice with Co60y model after model in the bone marrow using immunohistochemical methods on key genes ATM/ATR signaling pathway related protein and image analysis were analyzed; the removal of animal eye peripheral blood, their blood for blood analysis; testis after modeling, and ovary, using HE staining to observe pathological.Results:1. Immunohistochemical results:kidney group and blood group compared with the pathological aspects of the ATR protein expression differences (P<0.05) while the ATM gene in each group were not significantly different.2. HE staining of the reproductive system:testes extent of the damage kidney group compared with model group, the difference is larger, more obvious repair; blood and yin group degree of impairment less severe compared with model group; ovarian kidney group, blood group and the yin group to retain most of the outer granular cells, that some inner necrosis, shedding some of oocyte damage, extent of damage than those in model group.3. Peripheral Results:WBC, neutrophil, Kidney group and blood group and pathological group differences (P<0.05), lymphocytes of view, yin difference between group and pathological group (P<0.05) and hemoglobin of view, three groups and pathology group had significant difference (P<0.05).Conclusion:1. Bushentianjing be for ATR gene expression; 2. Bushentianjing, to a certain extent, the reproductive function of irradiated mice has a protective effect; 3. Bushentianjing side for bone marrow after radiation Cells have repair certain promotion, its more obvious for white blood cell proliferation. Summing Bushentianjing side with anti-radiation, protecting reproductive function in mice, and promote the growth of blood cells.
