菊米总黄酮的提取纯化及其降血压降血脂作用

【摘要】 菊米,为菊科植物野菊花(Chrysanthemum indium L.)的蕊杀青烘干而成。被中草药界誉为四大怀药之一,清代药学家赵学敏在《本草纲目拾遗》中早有记载,因其具有清热解毒、清火明目、平肝降压、心血管保健等作用,在浙江省作为一种饮品有着悠久的历史。本研究采用纤维素酶酶解结合超声波辅助提取,利用单因素试验和正交试验进行工艺优化,并通过大孔树脂层析,旨在分离纯化菊米中的黄酮类化合物;再以提取物为原料制备包衣微丸,试验结果表明其有效物质含量、含水量及性状在10个月内保持稳定;并用高效液相色谱法初步建立了菊米总黄酮的指纹图谱,为菊米制剂的质量控制提供了参考;采用自发性高血压大鼠模型和高脂血症大鼠模型,证实菊米总黄酮具有调节大鼠血压水平和高脂血症的作用,菊米总黄酮在保护心血管健康,治疗高血压高血脂等疾病方面尚有较大的研究价值。菊米作为浙江省重要的农副产品,纯化其功效成分,研制高附加值的保健食品,对经济发展和社会进步有重要意义。

【Abstract】 Jumi, the flower buds of Chrsanthe memindieum (Chrysanthemum indium L.), has been recorded in " Supplement to Compendium of Materia Medica" by pharmaceutist Zhao Xue-min in Qin Dynasty and used for a long time as Chinese traditional medicine in antibacterial, anti-inflammatory activities and to facilitate discharge of urine,protect liver, improve eyesight, decrease blood pressure and keep weight. In the present study, the extraction and purification of flavonoids from Jumi has been investigated. By the combination of ultrasonic and cellulase techniques we raised a novel and optimum method to isolate and purify successfully the flavonoids from Jumi by using of the chromatography with macroporous resin according to the single and orthogonal test; The Jumi extracts were used as raw material for extracting flavonoids prepared pellets, results showed that content of the bio-active compounds, water content and other physical characters had no obvious deterioration in 10 months; The HPLC fingerprint was established for quality assessment and control of Jumi extract and capsules; By using SHR and hyperlipidemic model rats, level of decreasing blood pressure (BP), heart rate (HR) and antiatheroscloresis ability of food-roune hyperlipidemia in rats by total flavonnoids from Jumi were observed in this study. Jumi is an important agricultural product in Zhejiang Province, therefore, the extraction and purification of its functional components and the development of health food products with high added value are of great significance for the development of economy and society.
