趋化因子受体CXCR3在乳腺癌中的表达及意义

【摘要】 目的研究趋化因子受体CXCR3在乳腺癌中的表达,分析其与浸润性乳腺癌相关临床病理指标之间的关系。方法应用RT-PCR和实时荧光定量PCR(Real-Time PCR)检测18例浸润性乳腺癌组织及其癌旁正常组织中CXCR3 mRNA的表达;应用免疫组织化学方法检测CXCR3在80例浸润性乳腺癌、20例乳腺纤维腺瘤和15例正常乳腺组织中的表达。结果(1)RT-PCR和实时荧光定量PCR结果显示乳腺癌组织中CXCR3 mRNA表达水平明显高于癌旁正常组织(P<0.05)。(2)免疫组化结果显示,CXCR3在乳腺癌组织中的表达明显高于其在乳腺纤维腺瘤和正常乳腺组织中的表达(P<0.05);乳腺癌组织中淋巴结转移阳性组CXCR3的表达明显高于淋巴结转移阴性组(P<0.05);CXCR3表达水平与乳腺癌患者淋巴结转移累及数目、肿瘤大小和pTNM分期呈显著正相关(P<0.05)。结论趋化因子受体CXCR3是一类参与乳腺癌病程进展的重要分子,其表达与多项临床病理指标,尤其是淋巴结转移密切相关,提示其可作为预测乳腺癌淋巴结转移及预后的免疫病理学指标。

【Abstract】 Objective To investigate the expression of CXCR3 and its association with clinicopathologic features in breast carcinoma.Methods The expression levels of CXCR3 in eighteen samples of breast cancer and corresponding normal tissues were investigated using reverse transcriptase-polymerase chain reaction (RT-PCR) analysis, real-time RT-PCR analysis.Immunohistochemistry was carried out to examine the expression of CXCR3 in 80 breast cancers,20 breast fibroadenoma and 15 normal breast tisues.Results (1) RT-PCR analysis showed higher levels of CXCR3 in breast cancer tissues than that in normal breast tissues. Real-time RT-PCR analysis showed that expression levels of CXCR3 in breast cancer tissues were significantly higher than that in the corresponding normal breast tissues(P<0.05). (2) Immunohistochemistry analysis showed that the positive rate of CXCR3 in breast cancer tissues was significantly higher than that in breast fibroadenoma and normal breast tisues(P<0.05). The expression levels of CXCR3 in lymph node-positive group was higher than that in lymph node—negative group(P<0.05); The expression of CXCR3 was positively correlated with the number of positive lymph node involved by metastasis, tumor size and pTNM tumor stage(P<0.05).Conclusions Chemokine receptor CXCR3 was up-regulated in breast cancer, and was associated with the progression of breast cancer; CXCR3 might be a novel molecular marker to predict lymph node metastasis and prognosis of breast cancer.
