痛泻要方半仿生提取法的研究

【摘要】 目的:优选痛泻要方半仿生提取工艺;进行该方半仿生提取液的质量控制初步研究。方法:以芍药苷、橙皮苷、升麻素苷、5-O-甲基维斯阿米醇苷的含量、白术特征峰面积、HPLC梯度洗脱总面积、干浸膏得率为指标,采用均匀设计优选痛泻要方半仿生提取的工艺条件;再以上述指标优选最佳醇沉浓度;在此基础上,作SBE法(半仿生提取法)、SBAE法(半仿生提取醇沉法)、WE法(水提法)、WAE法(水提醇沉法)4种方法提取液的成分比较;最后,用高效液相色谱法研究该方半仿生提取液中成分的含量测定和指纹图谱。结果:痛泻要方半仿生提取法的最佳工艺为:提取3次,水的pH值依次为为2.0、6.5、8.5;提取时间依次为60min、30min、30min;醇沉浓度以70%为佳;4种方法提取液成分的综合评价指标Y值的大小顺序为:SBE液>WE液>SBAE液>WAE液;该方半仿生提取液的HPLC指纹图谱有13个共有峰,4味药材各有其特征峰。结论:痛泻要方成分提取以SBE法为佳;该方半仿生提取液的HPLC指纹图谱及含量测定稳定可靠,可用于控制该方半仿生提取液的内在质量。

【Abstract】 Objective:To optimize the conditions of Tongxieyaofang Medicine by semi-bionic extraction (SBE). To study the quality control of Tongxieyaofang Medicine.Method:First, we optimize the best extraction conditions of the semi-bionic extraction for Tongxieyaofang Medicine with peoniflorin, prim-o-glucosylcimifugin, hesperidin, 5-O-methylvisamminol hilieidum,the characteristic peak area of Atractylodes macrocephala, the total integral area by HPLC, dried extract weight as the indexes. Then, with the indexes mentioned above, we optimize the best alcohol precipitation concentration. According to the former conditions, the four kinds of extract methods, including the semi-bionic extraction(SBE), the water extraction (WE), the semi-bionic extraction by precipitation with alcohol (SBAE) and the water extraction by precipitation with alcohol (WAE) were compared by the contents of the index ingredients.Furthermore we study the quality control of Tongxieyaofang Medicine.Result:The technological conditions of the semi-bionic extraction for Tongxieyaofang Medicine is pH in turn:2.0,6.5,8.5; Time:60min,30min,30min. the best alcohol precipitation concentration is 70%. The sequence of total index Y value of the four extraction technology is:YSBE> YWE >YSBAE>YWAE.The HPLC fingerprint of Tongxieyaofang Medicine have 13 peak.And anyone of 4 medicinal herb has its characteristic peak.Conclusion:SBE method is the best among the four methods in the extraction of components of Tongxieyaofang Medicine. It is stable and reliable that HPLC fingerprint of Tongxieyaofang Medicine.And we can use the HPLC fingerprint to control the quality of Tongxieyaofang Medicine.
