宫环止血汤对宫环出血肝郁血瘀证患者血清蛋白质表达的影响

【摘要】 目的：从血清蛋白质表达水平探讨宫环止血汤治疗宫环出血肝郁血瘀证的机理。方法：追踪采集宫环出血妇女置器前后和宫环止血片治疗前后血清,并采集置器后正常妇女的血清,确定95份有效血清,共分为五组,A组(置环前血清样本,置环后月经基本正常)22份,B组(置环前血清样本,置环后月经异常)9份,C组(置环后血清样本,置环后月经基本正常)13份,D组(置环后血清样本,置环后月经异常,肝郁血瘀证)41份,E组(对肝郁血瘀证宫环出血患者进行治疗后的血清样本)10份。采用蛋白质芯片-飞行质谱技术进行检测,得到血清蛋白质图谱。用Biomarker Wizard软件比较各血清蛋白质图谱,从宫环止血片治疗前后血清蛋白质表达水平上的变化,找出与治疗相关的蛋白质,结合生物信息,探讨其作用机理。结果：通过宫环止血汤治疗后,许多异常表达的血清蛋白含量得到了纠正,恢复了正常,质核比为22876.8和23480.9的二种血清蛋白与宫环止血汤的治疗机理有较大关联。结论：通过检索蛋白质数据库,找出Lin7b和Rab25可能与宫环止血汤的治疗机理有关,为下一步研究指明了方向。更多还原

【Abstract】 Objective:To deal with the treatment mechanism of Gonghuanzhixue soup treating metrorrhagia after women wearing IUD of liver depression and blood stasis syndrome from expression level of serum protein.Methods:Collecting abnormal metrorrhagia women’s serum of before and after inserting IUD,and before and after treatment using Gonghuanzhixue therapeutic,and the normal women’s serum after wearing IUD determining the effective serum 95 were divided into five groups, A group(serum samples pre-set IUD,normal menstruation after the set IUD)22,B group (serum samples pre-set IUD,abnormal menstruation after the set IUD)9,C group(serum samples after the set IUD,normal menstruation after the set IUD)13,D group(serum samples after the set IUD, abnormal menstruation after the set IUD,liver depression and blood stasis syndrome)41,E group(serum samples before and after treating metrorrhagia after women wearing IUD of liver depression and blood stasis syndrome) 10.Using SELDI-TOF-MS serum protein mass spectrums were obtained. Compared with the Biomarker Wizard software detecting serum protein mass spectrums, from Gonghuanzhixue soup before and after treatment of serum protein expression level changes,treatment-related proteins were identified, combined with bioinformatics,to explore its mechanism. Results:Through the treatment Gonghuanzhixue soup, many abnormal expression of the serum protein levels have been corrected and restored to normal.It was founded that charge-to-mass ratio were 22876.8 and 23480.9 were related to the treatment mechanism of Gonghuanzhixue soup.Conclusion:From retrieving the protein databases,Lin7b and Rab25 were possible related to the treatment mechanism of Gonghuanzhixue soup.
