超滤-纳滤联用用于干姜中指标性成分6-姜辣素的浓缩

【摘要】 目的:探索超滤-纳滤联用技术在干姜中指标性成分6-姜辣素浓缩中的应用。方法:以总蛋白和6-姜辣素透过率为指标,考察超滤过程。在单因素试验的基础上,采用Box-Behnken响应面法,以6-姜辣素的截留率为指标,考察滤膜截留相对分子质量(Mr)、6-姜辣素浓度和p H对纳滤过程的影响。结果:采用截留Mr为10 k Da的滤膜,对总蛋白的去除率为98.89%,6-姜辣素的透过率达到98.55%。纳滤过程最佳工艺参数为:截留Mr为350 Da、6-姜辣素质量浓度为150μg·ml-1、超滤液p H为7.0,对6-姜辣素的实际截留率为95.59%。结论:超滤-纳滤联用技术可有效完成对姜辣素类成分的浓缩,相比传统热浓缩优势明显。

【Abstract】 Objective: To apply ultrafiltration-nanofiltration in the concentration of indicator component 6-gingerol in Zingiberis Rhizoma. Methods: The ultrafiltration process was investigated by the permeability rate of total protein and 6-glyphosin. On the basis of single factor test,Box-Behnken response surface method was used to investigate the influences of molecular weight,6-gingerol concentration and p H value on the nanoflitration with the interception rate of 6-gingerol as the indicator. Results: The permeability rate of total protein was 98.89%,and the permeability rate of 6-gingerol was 98.55% by the membrane with interception molecular weight of 10 k Da. The optimal nanofiltration process parameters were as follows: the interception molecular weight was 350 Da,the 6-gingerol concentration was 150 μg·ml-1 and the p H value was 7.0,as a result,the actual interception rate of 6-gingerol was 95.59%. Conclusion: Ultrafiltration-nanofiltration coupling technology is more effective for alkaloids concentration than the traditional thermal concentration.
