菜豆再生体系及遗传转化体系研究进展

【摘要】 菜豆分子育种及功能基因组学分析需要高效稳定的再生体系和遗传转化体系。归纳了近年来国内外菜豆再生体系及遗传转化体系的研究进展。首先阐述了菜豆再生方法及研究现状,其中包括基于器官发生和体细胞胚发生不同途径菜豆再生体系的建立和相关的影响因素。其次概括了农杆菌介导法在菜豆遗传转化方面的研究进展,并分析了影响农杆菌介导遗传转化效率的主要因素,包括受体外植体类型和外植体损伤、农杆菌菌株及菌液浓度、共培养条件(时间、温度、光照时间)、乙酰丁香酮浓度等。然后叙述了有关基因枪介导菜豆遗传转化的研究进展。最后对目前菜豆再生体系和遗传转化体系存在的难题进行了总结,同时对今后研究的方向和思路做出了比较可靠的趋向预测,以期为建立高效稳定的菜豆再生体系和遗传转化体系提供参考依据。

【Abstract】 High-efficient and stable regeneration system and genetic transformation system are required for molecular breeding and functional genomics analysis of common bean. The regeneration system and Agrobacterium-mediated transformation of common beans at home and abroad in recent years were summarized. Firstly, the regeneration methods and research status of common bean were introduced, including the establishment of regeneration system based on organogenesis and somatic embryogenesis and related impact factors. Secondly, the research progress of Agrobacterium-mediated transformation in common bean was summarized, and the main factors affecting the efficiency of Agrobacterium-mediated transformation were analyzed, including the types of receptor explants and the damage of explants, Agrobacterium strain and bacterial concentration, co-culture conditions(time, temperature, the time of illumination), acetosyringone concentration, etc. Then the progress of gene gun-mediated transformation of common bean was reviewed.Finally, the problems in current regeneration system and genetic transformation system of common bean were summarized. At the same time, the future research directions and ideas were predicted, in order to provide references for the establishment of high efficient and stable regeneration system and genetic transformation system of common bean.
