忍冬不同药用部位活性成分的提取及含量分析

【摘要】 以河南封丘忍冬基地所产忍冬整株生药为研究对象,对比不同提取方法对忍冬各部位中活性成分含量大小的影响。用加热回流提取法、索氏提取法和超声波提取法分别对忍冬花蕾、叶、藤和根部中的活性成分进行提取,以绿原酸(CHA)、总黄酮作为指标成分,用薄层色谱法(TLC)、紫外色谱法(UV)和高效液相色谱法(HPLC)测定其含量。结果发现:与其他2种方法相比,用索氏提取法提取的有效成分最高,忍冬花蕾、叶、藤和根部中绿原酸质量分数分别为4.7%、3.0%、3.2%和2.1%,总黄酮的质量分数分别为6.9%、9.2%、3.6%和1.9%。提取效率从低到高分别是乙醇回流提取法、超声提取法和索氏提取法。3种提取方法以忍冬的花蕾、叶、藤、根部的粗提物中都检测到了绿原酸。绿原酸的含量从高到低为花蕾、叶、藤和根部;总黄酮的含量从高到低为叶、花蕾、藤和根部。本研究不仅通过对比得出了最优的提取方法,还为整株忍冬的综合开发利用提供了理论基础。

【Abstract】 We compared the effect of different extraction methods on the content of active ingredients in different medicinal parts of honeysuckle base in Fengqiu, Henan Province,to provide a basis for the comprehensive development of honeysuckle resources.We extracted active ingredients of flower buds,leaves,rattans and roots through reflux extraction,Soxhlet extraction and ultrasonic extraction,determined the contents of chlorogenic acid(CHA) and total flavonoids by TLC,UV and HPLC.Compared with the other two methods,Soxhlet extracted the highest active ingredients,and the content of CHA in honeysuckle buds,leaves,rattans and roots was 4.7%,3.0%,3.2% and 2.1%,respectively.And the content of total flavonoids was 6.9%,9.2%,3.6% and 1.9%, respectively.The extraction efficiency from low to high are ethanol reflux extraction,ultrasonic extraction and Soxhlet extraction.CHA was detected in the crude extract of flower buds,leaves,vine and roots of honeysuckle.The content of CHA was flower buds,leaves,rattans and roots.The total flavonoids content was leaves,flower buds,rattans and roots.The total flavonoids content was leaves,flower buds,rattans and roots.
