杜鹃红山茶的组织培养技术研究初探

【摘要】 杜鹃红山茶是一个极其珍稀的山茶品种,一度曾濒临灭绝.利用植物组织培养技术对其进行人工繁育研究具有重要的现实意义.研究了不同激素配比对杜鹃红山茶叶片和茎段诱导愈伤组织的影响.结果表明,杜鹃红山茶的叶片和茎段均能诱导出愈伤组织,其培养基中细胞分裂素和生长素配比1:1较为适宜诱导愈伤组织.适宜叶片诱导愈伤组织的培养基为MS+6-BA 1 mg/L+NAA 1 mg/L,诱导率为52.3%,适宜茎段诱导愈伤组织的培养基为MS+6-BA 1 mg/L+IBA 1 mg/L,诱导率为56.8%.另外,将带侧芽的茎段接于MS+6-BA 1 mg/L+IBA 0.01 mg/L+GA310 mg/L的培养基上培养30 d后观察,发现芽萌发良好,芽萌发率可达到57.1%.

【Abstract】 Camellia changii is an extremely precious variety of Camellia and endangered. Studies of the tissue culture technique were of great significance to the expand breeding of C.changii. The effects of different hormone combinations on callus induction of leaves and stems were studied in this paper. The results showed that leaves and stems of Camellia changii could induce callus. The suitable ratio of cytokinin/auxin for callus induction is 1∶1.The appropriate medium for leaves callus induction was MS +6-BA 1 mg/L +NAA 1 mg/L, the induction rate reached 52.3%, the appropriate medium for stems callus induction was MS+6-BA 1 mg/L+IBA 1 mg/L, and the induction rate reached 56.8%. In addition, when stems were cultured on the MS medium contained 6-BA 1 mg/L+IBA 0.01 mg/L+GA3 10 mg/L after 30 d, the buds was germinate good and the germination rate up to 57.1%.
