决明子多糖含量测定方法

【摘要】 本研究目的在于寻找最适合测定决明子多糖含量的方法,通过气相色谱法分析决明子多糖的单糖组成,根据其单糖摩尔比通过标准单糖来模拟决明子多糖,其测定结果与葡萄糖或甘露糖为标准单糖测定结果进行换算,以便更加准确测定决明子多糖的含量。从混标的线性关系看,苯酚硫酸法的相关系数比蒽酮硫酸法高(0.999 4 vs 0.996 8)。不同单糖标准的苯酚硫酸法测定决明子多糖的校正系数相对差异在合理范围,测得的结果也符合决明子多糖中甘露糖、混标、葡萄糖含量顺序,因此苯酚硫酸法更适合用来测定决明子多糖的含量。

【Abstract】 In this paper,the aim is to find the best method for determination of polysaccharide in Semen Cassiae.The monosaccharide composition of semen cassiae polysaccharide was analyzed by gas chromatography.According to the molar ratio of monosaccharide,the polysaccharide of Semen Cassiae was simulated by standard monosaccharide.The results were converted to glucose or mannose as standard monosaccharide in order to determine the content of polysaccharide of Semen Cassiae more accurately.From the linearity of the mixed standards,the correlation coefficient of Phenol-sulfuric acid method is higher than Anthrone-sulfuric acid method(0.999 4 vs 0.996 8).The relative difference of correction coefficient of polysaccharide in Semen Cassiae by different monosaccharide standard of phenol-sulfuric acid method is in the reasonable range.The result is also conform to the content of the order in Semen Cassiae that the highest contents of the mannose and the lowest contents of the glucose.Therefore,Phenol-sulfuric acid is more suitable for the determination of the contents of polysaccharide in Semen Cassiae.
