共生菌对荷瘤小鼠肺泡巨噬细胞基因表达的调控作用

【摘要】 目的探讨共生菌群对荷瘤小鼠肺泡巨噬细胞(AM)基因表达的调控作用。方法分离纯化共生菌缺失的荷瘤小鼠及共生菌正常的荷瘤小鼠AM,利用基因芯片技术筛选差异表达的基因并进行生物信息学分析。结果分选后的AM细胞纯度可达98%,共生菌缺失荷瘤小鼠与正常荷瘤小鼠相比较,AM细胞中差异性表达基因共有353个(变化倍数≥2),上调基因171个,下调基因182个;差异表达基因的基因分类(GO)主要为细胞凋亡、细胞增殖、细胞分化、信号传导、血管生成等;KEGG通路主要为氮素代谢、趋化因子信号、TGF-β信号及肿瘤通路等;M2型巨噬细胞特异性基因CCL24、Arg1、Retnla及IL-13等在共生菌缺失荷瘤小鼠AM细胞中表达上调。结论共生菌可以调控荷瘤小鼠AM细胞的基因表达,包括CCL24、Arg1、Retnla及IL-13等基因,差异表达基因与多个GO分类和KEGG通路密切相关。

【Abstract】 Objective To explore the regulation of the microbiota on the gene expression in alveolar macrophages(AM) from tumor-bearing mice.Methods AM were separated and purified from the tumor-bearing mice that have the commensal bacteria depleted or not,Microarray technology was used to screen the genes with different expression levels and the bioinformatics methods were used to analyze the differences.Results The purity of the separated AM was 98%.In the AM from the tumor-bearing mice with commensal bacteria depleted,compared with the ones with the intact commensal bacteria,there were 353 differentially expressed genes with the fold changes greater than 2 171 of them were upregulated and 182 of them were downregulated;the gene ontology(GO) of these differentially expressed genes were mainly cell apoptosis,cell proliferation,cell differentiation,signal transduction and angiogenesis etc.;their KEGG pathways are mainly nitrogen metabolism,chemokine signaling pathway,TGF-β signaling pathway and the pathway in tumor etc.;M2 macrophage specific genes,such as CCL24,Arg1,Retnla and IL-13 etc.,were upregulated in the AM from the mice with the depleted commensal bacteria.Conclusion The microbiota regulates the expression of the genes in AM from tumor-bearing mice,including CCL24,Arg1,Retnla and IL-13 etc.,those differentially expressed genes are associated with many GO process and KEGG pathways.
