新疆地区淡水贝类的形态学及分子分类研究
Morphological and Molecular Classification of Freshwater Mollusks in Xinjiang
【摘要】 为了解新疆地区淡水贝类种类及地理分布情况,采用形态学与分子生物学相结合的方法对新疆12个地区采集的淡水贝类进行分类鉴定。利用基于线粒体DNA (mt DNA)COI基因、核糖体内转录间隔序列2(ITS2)基因的DNA条形码技术进行PCR扩增、克隆及测序,对其进行分子分类鉴定。结果发现,21种淡水贝类主要隶属于腹足纲(Gastropoda)和双壳纲(Bivalvia),共4目、10科、14属;其中隶属双壳纲的贝类仅有1科1种;隶属于腹足纲的淡水螺占9科、13属,分属田螺科1种、豆螺科1种、扁卷螺科2种、椎实螺科7种、膀胱螺科2种、巴蜗牛科1种、坚齿螺科4种、琥珀螺科1种、嗜石螺科1种。
【Abstract】 In order to investigate the geographical distribution and species of freshwater mollusks, morphological, anatomical and molecular biological techniques were used to identify and classify freshwater mollusks collected from 12 areas of Xinjiang. COI and ITS2 gene fragments were amplified respectively by PCR with specific primers, cloned and sequenced. The molecular identification and phylogenetic analysis were performed. Morphological and molecular biological identification suggested that nearly 21 kinds of freshwater mollusks were collected during the survey and a total of 21 important species of molluscs in Xinjiang had been identified, which belongs to 4 orders, 10 families, 14 genus, which belongs to gastropoda and freshwater bivalve mollusks(Bivalvia), respectively. Only one family, one species of shellfish belongs to the bivalvia. Moreover, freshwater snails belongs to gastropods accounted for 9 families and 13 genera in which there was one species of Viviparidae,Bithyniidae,Bradybaenidae,Succineidae,and Hygromiidae and Unionidae,2 Species of Planorbidae and Physidae,7 Species of Lymnaeidae,4 Species of Camaenidae.
