七叶一枝花有效成分的提取工艺优化

【摘要】 采用回流提取法提取七叶一枝花根茎的主要有效成分重楼皂苷,用高氯酸显色法进行含量测定.设计正交试验,探讨了溶剂浓度、提取温度和提取时间对总皂苷的影响及最佳提取工艺.结果表明:影响重楼总皂苷得率的因素依次为提取温度>溶剂浓度>提取时间,最优提取条件是乙醇浓度75%、提取温度90℃、提取时间1.5 h.在此条件下,重楼总皂苷得率为12.74%.实验通过精密度、稳定性以及加样回收等方法学研究进行验证,RSD均小于2%.

【Abstract】 The main effective components of rhizome of Paris polyphylla were extracted by reflux extraction method,and the content was determined by perchloric acid chromogenic method. The effects of solvent concentration,extraction temperature and extraction time on total saponins were studied by orthogonal design. The results showed that the factors affecting the yield of total saponins from heavy floor was extraction time>extraction temperature>solvent concentration,the optimal extraction conditions is 75% ethanol concentration, extraction temperature 90 ℃, the extracting time 1.5 h.Under these conditions,the yield of total saponins was 12.74%. The experiments were verified by precision,stability and sampling recovery,and the results showed that RSD was less than 2%.
