大球盖菇液体菌种培养基碳氮源及培养条件的优化

【摘要】 为了促进大球盖菇液体菌种扩繁,采用单因素方差分析和正交试验的方法,对菌体培养的碳源、氮源和培养条件进行优化,通过测定大球盖菇菌丝球生物量指标,研究不同碳氮源和不同培养条件对大球盖菇生长的影响。结果表明:大球盖菇最适培养基为葡萄糖20 g·L-1,玉米粉10 g·L-1,麸皮5 g·L-1,KH2PO4 3 g·L-1,MgSO4 0.2 g·L-1。优化后,大球盖菇菌丝体生物量增加了174%,最佳培养条件是pH6,转速120 r·min-1及培养温度25℃。培养基和培养条件的优化可以显著促进大球盖菇菌丝的生长。

【Abstract】 In order to promote the expansion and propagation of the liquid strains of Stropharia rugosoannulata,this paper optimized carbon source,nitrogen source and culture conditions with the one-way analysis of variance and orthogonal experimental design,through the determination of Stropharia rugosoannulata mycelia biomass index,researched effect of the different carbon source,nitrogen source and different culture conditions on Stropharia rugosoannulata mycelial growth.The results showed that the optimal medium was 20 g·L-1 glucose,10 g·L-1 maize flour,5 g·L-1 bran,3 g·L-1 KH2PO4 and MgSO40.2 g·L-1.After optimization,mycelium biomass increased by 174%,the best culture conditions were pH6,rotating speed 120 r·min-1 and the temperature at 25℃.The optimization of culture medium and culture conditions could significantly promote mycelial growth of the Stropharia rugosoannulata.
