红树林菌株对毛用活性红染料的降解和脱色研究

【摘要】 通过梯度驯化,从土壤中分离筛选出能以毛用活性红PW-6g为唯一碳源的4株菌株,对其进行偶氮还原酶、苯胺双加氧酶、苯甲酸-1,2-双加氧酶、邻苯二酚-1,2-双加氧酶、邻苯二酚-2,3-双加氧酶活性测定,确定综合酶活性较好的菌株,鉴定其为Bacillus subtilis spp.菌株。对该菌株的脱色反应条件进行研究,结果表明,当PW-6g初始质量浓度为600 mg/L时,在pH为7.5、温度为35℃、接种量为4%、盐质量浓度小于40 g/L的条件下,Mabhk-3脱色48 h对PW-6g的脱色降解率可接近100%。通过HPLC分析,均检测不出脱色液中含有苯二胺、苯胺、邻苯二甲酸、苯甲酸和邻苯二酚,达到排放要求。

【Abstract】 Through gradient acclination,four strains that could use a kind of lanasol active red dye(PW-6 g) as sole carbon source were screened from natural mangrove soil. Of them,the activities of azoreductase,aniline dioxygenase,benzoate-1,2-dioxygenase,catechol-1,2-dioxygenase and catechol-2,3-dioxygenase were measured. Based on the prominent comprehensive enzymes activities,the strain was selected and identified as Bacillus subtilis spp.. The results from the study showed that the optimum initial concentration of PW-6 g was 600 mg/L,the optimum temperature for decolorization was 35 ℃,optimum pH was 7.5,optimum inoculating dose was 4%,and salt concentration was less than 40 g/L. Under the optimum conditions,the decolorization and degradation rate was nearly 100% after the treatment of PW-6 g with the strain Mabhk-3. It was found that p-phenylenediamine,aniline,phthalic acid,benzoic acid and catechol in the decolorization solution can not be detected by HPLC analysis and the decolorization solution met the emission specifications.
