铁皮石斛组织培养研究
Tissue Culture of Dendrobium candidum
【摘要】 铁皮石斛是一种名贵的中药材,药效显著,传统种植方法繁殖率低,加上人们掠夺式的采挖,导致其野生资源濒临枯竭。以幼嫩的铁皮石斛带节茎段为材料,对不同激素浓度、配比及添加不同种类的植物营养液对铁皮石斛组织培养的影响进行了研究。结果表明:铁皮石斛嫩茎的最佳诱导培养基为:MS+6-BA1.5 mg·L-1+NAA0.5 mg·L-1+蔗糖30 g·L-1,继代培养基可以用MS+6-BA1.5 mg·L-1+NAA0.5 mg·L-1+马铃薯汁100 g·L-1+活性炭1 g·L-1+蔗糖30 g·L-1,最佳的生根培养基为1/2MS+NAA0.3 mg·L-1+香蕉汁30 g·L-1+蔗糖30 g·L-1。为铁皮石斛的快速繁殖和工厂化育苗提供技术支撑。
【Abstract】 Dendrobium candidum is a valuable Chinese medicinal material with remarkable efficacy. The traditional breeding method has a low reproduction rate. Coupled with the predatory mining, the wild Dendrobium resources are on the verge of exhaustion. The effects of different hormone concentrations, ratios and different plant nutrient solutions on the tissue culture of Dendrobium candidum were studied with the young stems of Dendrobium candidum. Result shows that the optimal induction medium of Dendrobium candidum is: MS+6-BA 1.5 mg·L-1 +NAA 0.5 mg·L-1+sucrose 30 g·L-1, and the subculture medium can be MS+6-BA1.5 mg·L-1+NAA0.5 mg·L-1+ potato juice 100 g ·L-1+ C 1 g ·L-1+ sucrose 30 g· L-1, the best rooting medium is 1/2 MS+NAA 0.3 mg· L-1+ banana juice 30 g ·L-1+sucrose 30 g ·L-1. It can provide technical support for rapid propagation research and factory breeding of Dendrobium candidum.
