血府逐瘀汤抗大鼠心肌缺血损伤及其作用机制的实验研究

【摘要】 目的研究血府逐瘀汤抗心肌缺血损伤的作用及可能的作用途径。方法将大鼠随机分为假手术组、模型组、血府逐瘀汤组和地奥心血康组4组。采用结扎冠状动脉左前降支60min建立心肌缺血损伤模型。运用生物化学、免疫组化等实验技术,观察大鼠心肌缺血损伤后超氧化物歧化酶(SOD)、丙二醛(MDA)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、细胞凋亡、凋亡调控基因(Bax、Bcl-2)、细胞间粘附分子(ICAM-1)、半胱氨酸天门冬氨酸蛋白酶-3(Caspase-3)含量的变化,以及血府逐瘀汤对上述指标的影响。结果(1)血府逐瘀汤可显著降低心肌缺血损伤大鼠血清和心肌组织MDA含量(P<0.05),明显提高SOD活性(P<0.05)。(2)血府逐瘀汤可显著降低心肌缺血损伤大鼠血清LDH和CK含量(P<0.01,P<0.05)。(3)血府逐瘀汤可显著降低心肌缺血损伤大鼠心肌凋亡细胞数量(P<0.01)。(4)血府逐瘀汤可明显抑制心肌缺血损伤大鼠心肌Bax表达(P<0.01),显著促进心肌Bcl-2表达(P<0.05)。(5)血府逐瘀汤可显著降低心肌缺血损伤大鼠心肌ICAM-1和Caspase-3的表达(P<0.05,P<0.01)。结论氧自由基、细胞凋亡、细胞粘附分子、半胱氨酸天门冬氨酸蛋白酶参与心肌缺血损伤的发生。血府逐瘀汤具有抗心肌缺血损伤的作用,这一作用可能是通过以下途径实现的:减轻氧自由基损伤;减少酶的外漏;减少心肌细胞凋亡的数量;抑制心肌Bax表达、促进Bcl-2表达;抑制心肌ICAM-1和Caspase-3的表达。总之,血府逐瘀汤可从多个环节减轻心肌缺血损伤,达到其抗心肌缺血损伤的目的。

【Abstract】 Objective To study the effect of xiefu zhuyu decoction against myocardial ischemia injury and the possible functional pathways. Methods Rats were divided into four groups:sham operating group, model group,xiefu zhuyu decoction group,diao xinxie kang goup.The model of myocardial ischemia injury was produced by ligaturing anterior descending branch of left coronary artery for 60 minutes in rats.The contents of SOD、MDA、LDH、CK,apoptosis,expression of Bax、Bcl-2、ICAM-1 and Caspase-3 after ischemia and the effects on above items of xiefu zhuyu decoction were observed.Results(1 ) Xiefu zhuyu decoction could significantly reduce the content of MDA in ischemia injury myocardium and blood of rats(P<0.05 ),while obviously increasing the activity of SOD in ischemia injury myocardium and blood of rats(P<0.05 ). (2 ) Xiefu zhuyu decoction could significantly decrease the levels of CK and LDH in the rats blood of myocardial ischemia injury(P<0.01,P<0.05 ). (3) Xiefu zhuyu decoction could significantly reduce the number of myocardial cell apoptosis(P<0.01)(4) Xiefu zhuyu decoction could obviously reduce the expression of Bax(P<0.01),while significantly promoting the expression of Bcl-2 in ischemia myocardium(P<0.05 ).(5 ) Xiefu zhuyu decoction could significantly reduce the expression of ICAM- 1 and Caspase-3 in ischemia myocardium(P<0.05,P<0.01).Conclusion Oxygen free radical,enzyme,apoptosis,cell adhesion molecule,cysteinyl aspartate specific proteinase participate myocardial ischemia injury.Xiefu zhuyu decoction has the effect against myocardial ischemia injury.This effect of Xiefu zhuyu decoction could be carried out by the following pathways:reduce the injury by oxyen free radial,decrease the out-leaking of enzyma,decrease the number of myocardial cell apoptosis,reduce the expression of Bax,、ICAM-1 and Caspase-3,improve the expression of Bcl-2. By the above ways,xiefu zhuyu decoction could effectively prevent the myocardial ischemia injury.
