【摘要】 目的：通过观察益气养阴、活血化瘀方药对柯萨奇B_3m诱导慢性心肌损伤小鼠心肌组织ADAMTS-1、TGF-β₁的影响，探讨其作用机制，为早期防治病毒性心肌病提供客观依据。方法：采取多次反复腹腔接种CVB_3m方法诱导Balb／c小鼠发生慢性心肌损伤，建立早期病毒性扩张型心肌病动物模型。210只Balb／c小鼠，随机分为4组，空白对照组3 0只，模型对照组、中药高剂量组、中药低剂量组各60只。采用HE、VG染色观察其心肌病理形态学及超微结构的变化；采用RT-PCR法观察心肌组织ADAMTS-1和TGF-β₁表达水平的变化。结果：1．益气养阴、活血化瘀方药能降低病毒性心肌病小鼠心脏质量指数，以中药高剂量组效果为佳。2．益气养阴、活血化瘀方药能明显改善病毒性心肌病小鼠心肌病理形态及超微结构的变化，从而减轻心肌病理损伤。3．益气养阴、活血化瘀方药能下调病毒性心肌病小鼠心肌组织ADAMTS-1的表达水平，以中药高剂量组效果为佳。4．益气养阴、活血化瘀方药能下调病毒性心肌病小鼠心肌组织TGF-β₁的表达水平，以中药高剂量组效果为佳。结论：益气养阴、活血化瘀方药能减轻病毒性心肌病小鼠的病理损伤，下调心肌组织ADAMTS-1和TGF-β₁的表达，这可能是其防治病毒性心肌病的作用机制之一。更多还原

【Abstract】 Objective:To observe the effects the formula of supplementing qiand nourishing yin,promoting blood circulation to dispel blood stasis onthe thecoxB3m induce chronic myocardial injuring the rat’myocardiumtissue ADAMTS-1、TGF-β₁ model with viral cardiomyopathy.To investigateits mechanics,then to provide academic warranties for preventing andcuring the viral myocardial disease early.Methods:Establishing the animal models of the early viral dilatedcardiomyopathy with the method of repeating injecting CVB3m virus to theperitoneal cavity of the Balb/c rats.Deviding the 210 Balb/c rats into fourgroups random.The blank group has 30 rats.The model group,the high dosechinese traditional medicine group and the low dose chinese traditionalmedicine group have 60 rats each.mand,the ultrastructure of the formula ofHE、VG c hromosome dye and the expression of the TNF-α、mRNA in themuscletissue of the heart.To bserve the express of ADAMTS-1 and TGF-β₁in the myocardial tissue with the forual of RT-PCR.Result:1.The formula of supplementing qi and nourishing yin,promotingblood circulation to dispel blood stasis can depress the quality indicatorof the rat model with viral cardiomyopathy.And the effects of the high dosechinese traditional medicine group is the best.2.The formula of supplementing qi and nourishing yin,promotingblood circulation to dispel blood stasis can improve the changes of thepathomorphismand and the ultrastructure of the rat model with viralcardiomyopathy,then to reduce the injury on its myocardial pathology.3.The formula of supplementing qi and nourishing yin,promotingblood circulation to dispel blood stasis can play down the expression of theADAMTS-1 Level of the rat model with viral cardiomyopathy.And theeffects of the high dose chinese traditional medicine group is the best. 4.The formula of supplementing qi and nourishing yin,promotingblood circulation to dispel blood stasis can play down the expression of theTGF-β₁ Level of the rat model with viral cardiomyopathy.And the effects ofthe high dose chinese traditional medicine group is the best.Conclusions:The formula of supplementing qi and nourishing yin,promoting blood circulation to dispel blood stasis can alleviate thepathologic injury,and lowing the expression of myocardial tissueADAMTS-land TGF-β₁.and then to allieviate the bisintigrin resolve outsidethe cells and myocardial restructure.This may be its function mechanism ofits preventing and curing vial myocardial disease.更多还原
