地乌和蛇莓的化学成分研究

【摘要】 地乌Anemone flaccida Fr.Schmidt,为银莲花属植物的干燥根茎,具有祛风湿,助筋骨,治疗风湿疼痛,跌打损伤等功效。本文运用各种分离技术从地乌中分离鉴定了17个化合物的结构,其中化合物oleanolic acid 3-O-β-D-glucopyranosyl-(6’-butyryl)28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl为新化合物,化合物Monoethyl Malonate为新天然产物。其中的7个化合物:水合氧化前胡内酯、齐墩果酸3-O-β-D-吡喃葡萄糖-(1→2)-α-L-吡喃阿拉伯糖苷,齐墩果酸3-O-β-D-吡喃葡萄糖醛酸,齐墩果酸3-O-β-D-吡喃葡萄糖醛酸甲酯,齐墩果酸28-O-α-L-吡喃鼠李糖-(1→4)-β-D-吡喃葡萄糖-(1→6)-β-D-吡喃葡萄糖苷,齐墩果酸3-O-β-D-吡喃葡萄糖醛酸甲酯28-O-α-L-吡喃鼠李糖-(1→4)-β-D-吡喃葡萄糖-(1→6)-β-D-吡喃葡萄糖苷,齐墩果酸3-O-α-L-吡喃阿拉伯糖-(1→2)-α-L-吡喃鼠李糖28-O-α-L-吡喃鼠李糖-(1→4)-β-D-吡喃葡萄糖-(1→6)-β-D-吡喃葡萄糖苷,为首次从地乌植物中分离得到。以HeLa、BEL-7402、HL-60细胞作为药理活性研究对象,考察了地乌中分离得到单体化合物的抗肿瘤作用,实验结果表明三萜皂苷类化合物对HL-60细胞的抑制作用较强,对BEL-7402细胞的抑制作用相对较弱,新化合物1对上述细胞的生长具有较强的抑制作用。中药蛇莓Duchesnea indica(Andr.)Focke.为蛇莓的全草,具有清热、凉血、消肿、解毒的功效,民间多用于治疗热病、咳嗽等疾病。本文运用各种分离技术,对乙酸乙酯提取物进行了化学成分研究,从中分离鉴定了10个化合物,其中3个乌苏烷型三萜化合物中:新化合物18具有三元氧环结构,化合物19、20含有两个双键,这在五环三萜的天然产物中较为少见。

【Abstract】 Anemone flaccida Fr.Schmidt has been used for the treatment of arthralgia, headache and cold as a folk medicinal herb.By column chromatography,seventeen compounds were isolated from the EtOH extract of Anemone flaccida and elucidated on the basis of spectroscopic methods,including one new compound:oleanolic acid 3-O-β-D-glucopyranosyl-(6’-butyryl)28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl -(1→6)-β-D-glucopyranosyl,one new natral product:Monoethyl Malonate.And seven compounds,oleanolic acid 3-O-β-D-glucopyranosyl-(1→2)-α-L -arabinofurnoside,oleanolic acid 3-O-β-D-glucuronopyranose,oleanolic acid 3-O-β-D-glucuronopyranose methyl ester,oleanolic acid 28-O-α-L-rhamnopyranosyl (1→4)-β-D-glucopyranosyl(1→6)-β-D-glucopyranosyl and so on, were isolated from this plant for the first time.The isolated compounds were screened by antitumor bioassay.New compound(1) revealed significant antitumor activities to Hela and HL-60.Duchesnea indica(Andr.) Focke.is one of important medicinal plants used for anti-tumor,anti-bacterial,anti-inflammatory,immunity-improving.By column chromatography,ten compounds were isolated from the EtOH extract of Duchesnea indica and elucidated on the basis of spectroscopic methods,including one new compounds:2α-3β-dihydroxyurs- 12-en- 18,19-epoxy-27-oic acid,and others compounds were 2α-3α-dihydroxyurs- 12-en-27-oic acid,2α-3α-dihydroxyurs- 12-en-27 -oic acid,3-hydroxy-4-methoxy-trans-cinnacic acid,4-hydroxy-trans-cinnacic acid were isolated from this plant for the first time.
