海藻酸钠—明胶共混体系为载体的可注射式组织工程骨的研究

【摘要】 目的:探讨海藻酸钠—明胶共混体系为载体的可注射式组织工程骨的理化性能、生物相容性及其在临床上的应用前景。方法:从兔骨髓中分离骨髓间充质干细胞,进行体外培养、分离和扩增,诱导向成骨细胞分化并进行检测和观察。探讨了不同浓度的氯化钙溶液对成骨细胞的影响作用。研究不同配比共混体系的理化性能和不同消毒方法对其的影响作用和成骨细胞在体系中的生物学行为。在从细胞遗传学角度考察了共混体系安全性的基础上,进行动物实验,观察材料的组织相容性和不同时间段新骨的形成。结果:骨髓间充质干细胞经诱导培养后分化为成骨细胞,Ca2+浓度对成骨细胞的代谢活性有影响作用。共混体系理化性能随不同配比的浓度、温度有所变化。各种消毒方法对其理化性能均有影响,其中,以高压蒸汽法对海藻酸钠粉末的影响最小。成骨细胞在共混体系中,呈悬浮状生长并增殖。植入共混体系/成骨细胞凝胶的动物,未见到有染色体型畸变。凝胶注射植入实验兔皮下后有软骨样组织和骨组织形成,并以软骨化骨的形式修复了实验动物的颅骨极限缺损。结论:骨髓间充质干细胞可通过体外诱导培养分化为成骨细胞。不同浓度和作用时间的氯化钙对成骨细胞具有影响作用。海藻酸钠—明胶共混体系的理化性能可满足作为成骨细胞载体的需要。高压蒸汽对海藻酸钠粉末进行消毒的方法对共混体系的理化性能影响最小。共混体系无明显致染色体型畸变作用。通过注射方式在动物软组织中有异位成骨作用。可以用来进行骨缺损的修复。

【Abstract】 Object To explore physico-chemical property,biocompatibility and clinical application of injectable tissue engineering bone carried by algin-gelatin complex.Methods Bone marrow mesenchymal stem cells were abstracted from rabbit bone marrow.And then culture,isolation and amplification were performed in vitro. Osteoblasts,drived from Bone marrow mesenchymal stem cells,were measured and observed.Effect of different concentration solution of calcium chloride on osteoblasts was studied.Effect of different matching ratio complex and sterilization methods on complex was investigated.From cytogenetics,safety,and biocompatibility,of complex, and duration of osteogenesis were explored.Results Bone marrow mesenchymal stem cells were induced into osteoblasts. Concentration of Ca2+ could influence metabolic activity of osteoblasts.Different matching ratio,temperature and sterilization methods could change physico-chemicai property of complex.Vapor sterilization for powder of algin had little effect on physico-chemical property of complex.Osteoblasts in complex system grew and proliferated in suspension.To the animals,which were transplanted in complex system of gels and cells,aberration of chromosomal pattern did not occur.After transplantation of the gel by injected into subcutaneous tissue,chondrogenesis and osteogenesis happened.And maximum defect of skull was repaired with os cartilaginea.Conclusion Bone marrow mesenchymal stem cells can be induced into osteoblasts in vitro.Different concentration and duration of calcium chloride have effects on osteoblasts.Physico-chemical property of algin-gelatin complex can satisfy requirement of osteoblasts cartier.Vapor sterilization for powder of algin has little effect on physico-chemical property of complex.Complex system has no effect of chromosomal pattern aberration.Using the injectable method heterotopic bone can be applied to repair defect of bone.
