祛斑汤治疗黄褐斑的作用及机理探讨

【摘要】 目的:黄褐斑是一种获得性色素沉着性皮肤病,其发病机理复杂,现代医学对其研究尚未完全明了,目前无满意的疗法,中药以其副作用小,疗效持久显示出一定的优势。本实验研究是为了揭示祛斑汤治疗黄褐斑的作用机理,为该方在临床上治疗本病提供理论依据。方法:体外采用多巴色素法测定祛斑汤对酪氨酸酶(TYR)活性的抑制作用,并且研究了祛斑汤抗多巴自动氧化、抗紫外线催化及促黑素分解作用;以A375人黑素瘤细胞为受试细胞,应用MTT法、多巴氧化法及氢氧化钠裂解法测定了祛斑汤及其含药血清对细胞增殖、TYR活性及黑素合成的影响;采用肌肉注射黄体酮复制黄褐斑小鼠模型,观察了祛斑汤对肝脏、皮肤组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)的影响,同时通过HE染色、免疫组化方法观察了对皮肤黑素细胞(MC)的影响。结果:祛斑汤能抑制TYR活性,并具有抗多巴自动氧化及抗紫外线催化作用。祛斑汤及其含药血清对体外培养的A375人黑素瘤细胞具有抑制细胞增殖,降低TYR活性和黑素含量作用。祛斑汤能升高黄褐斑小鼠皮肤和肝脏中SOD的活性,降低MDA的含量,同时能明显使MC生成减少、抑制黑素合成,从而减轻黄褐斑小鼠皮肤色素沉着。结论:通过上述实验,初步揭示了祛斑汤治疗黄褐斑的作用机理,其机制可能是抑制MC增殖,降低TYR活性,抗氧化等途径。

【Abstract】 Objective:Chloasma is an acquired pigmentation dermatosis.It has a complex mechanism.West medicine hasn’t studied it completely and obviously.There is no satisfying treatment in chloasma now.Chinese medicine shows some superiority with its less side-effects and lasting curative effect.The aim of this study is to reveal the mechanisms of Quban decoction to cure chloasma.It can provide a theory basis for its clinical application on Chloasma.Methods:The inhibition of Quban decoction on tyrosinase was determined by the dopachrome method in vitro,and Quban decoction was investigated by anti-DOPA autoxidation activity,anti-ultraviolet catalysis activity,and promoting degradation of melanin.A375 human melanoma cell was employed in this study to investigate the effects of Quban decoction and Medicated serum of its on cellular proliferation, tyrosinase activity and melanin content.The model of chloasma in female mice was induced by musculal injection of progesterone for one month,the effects of Quban decoction on SOD,MDA and NO in skin and liver were detected,and skin melanocyte was observed.At the same time,samples were taken to store in the fixation liquid for HE and immunohistochemical performance.Results:The results showed that Quban decoction could inhibit the activity of tyrosinase,and at the same time it showed anti-DOPA autoxidation activity and anti-ultraviolet catalysis activity.Quban decoction and Medicated serum of its had the effects on inhibiting the proliferation,and decreasing tyrosinase activity and melanin content of A375 human melanoma cell in vitro.Quban decoction could increase the activity of SOD,while the contents of MDA were obviously decreased in skin and liver,and at the same time a decrease of melanin production in the mice model was observed.Conclusions:Based on the above experiments,we revealed the mechanisms of Quban decoction to cure chloasma,and its possible mechanisms may be related to inhibiting melanocyte roliferation,decreasing tyrosinase activity,and anti-oxidization, and so on.
