中药山慈菇的化学成分及其抗肿瘤活性研究

【摘要】 山慈菇(Pleione bulbocodioides(Franch.)为中国药典收载的少常用中药。其味甘、微辛、性凉,归肝、脾经,具有清热解毒、化痰散结的功效,主要用于痈肿疔毒、瘰疬,蛇虫咬伤等症,其化学成分的研究较少,本文从山慈菇中分离得到60个单体化合物,通过1H-NMR,13C-NMR,2D-NMR,高分辩质谱等波谱分析方法鉴定了其中30个化合物的结构,其中6个为新化合物,分别为: 4-(4’’–hydroxy benzyl)-3-(3’-hydroxyphenethyl)furan-2(5H)-one.(1),3-(3’-hydroxy phenethyl)furan -2(5H)-one(2),2-(4’’-hydroxybenzyl)-3-(3’–hydroxy phenethyl)-5 -methoxycyclohexa -2,5 -diene-1,4-dione(3),(3 -hydroxy -9-(4’-hydroxy-3’-methoxy phenyl)-11-methoxy-5, 6, 9, 10-tetrahydro phenanthro [2, 3-b] furan -10-yl) methyl acetate (4),(7-hydroxy-2 -(4-hydroxy-3-methoxy phenyl)-10-methoxy-2,3,4,5 -tetrahydrophenanthro [2,1-b] furan-3-yl)methyl acetate(5),(3-hydroxy-9-(4’- hydroxy -3’-methoxyphenyl)-11-methoxy -5, 6, 9, 10–tetrahydro phenanthro [2, 3-b] furan -10-yl) methyl acetate(6);18个为首次从该植物中分离得到,分别为:gymconopin D(10), 3, 5-diimethoxy-3’-hydroxy- bibenzyl(12), Kayaflavone(15), 5, 5’’, 7, 4’, 4’’’, 7’’-六羟基-[3’-8’’]双黄酮(16), 4-羟基苯乙酸甲酯(17),对羟基苯甲醛(18),对羟基苯甲酸(19), 4-(methox- ymethyl)phenol(20),对羟基苯丙酸甲酯(21),间羟基苯丙酸甲酯(22),肉桂酸(23),间羟基苯甲酸(24), 4-(ethoxymethyl) phenol(25), 4-Oxopentanoic acid(26), 5-羟甲基糠醛(27), 3-羟基苯丙酸(28),对苯二酚(29),天麻苷(30)。本文还对山慈菇石油醚提取物,乙酸乙酯提取物,正丁醇提取物及部分单体做了体外抗LA795细胞活性筛选,结果表明:在粗提物中石油醚萃取物和乙酸乙酯萃取物有一定活性,各单体化合物中4,7,12,13,29有一定活性,本研究对深入研究和开发中药山慈菇提供了参考。

【Abstract】 Pleione bulbocodioides is sweet, pungent in taste and cool in nature and its therapeutical action is related to the chanels of liver and spleen. It has actions of removing heat and counteracting toxicity, dissipating phlegm and eliminating stagnation. It is applied in the treatment of edema, crewels, snake and insert bite. The chemical investigation on ethanol extract of the tubers of P. bulbocodioides led to the isolation and identification of 6 new compounds together with 24 known compounds in which 18 compounds were obtained from the titled plant for the first time. Their structures were elucidated on the basis of 1H-NMR, 13C-NMR and 2D-NMR spectral data, respectively. The six new compounds were determined as follows:: 4-(4’’– hydroxy benzyl)-3-(3’-hydroxyphenethyl)furan- 2(5H)- one .(1),3-(3’- hydroxy phenethyl) furan -2(5H)-one (2),2-(4’’-hydroxybenzyl)-3-(3’–hydroxy phenethyl) -5-methoxy cyclohexa -2, 5 -diene-1,4-dione (3),(3 -hydroxy -9-(4’-hydroxy -3’-methoxyphenyl)-11-methoxy-5, 6, 9, 10-tetrahydro phenanthro [2, 3-b] furan -10-yl) methyl acetate (4),(7-hydroxy-2 -(4-hydroxy- 3-methoxy phenyl)-10-methoxy -2, 3, 4, 5-tetrahydro phenanthro [2,1-b] furan-3-yl)methyl acetate (5),(3-hydroxy-9- (4’-hydroxy -3’-methoxyphenyl)-11-methoxy -5, 6, 9, 10–tetrahydro phenanthro [2, 3-b] furan -10-yl) methyl acetate (6).. The 18 compounds obtained from the titled plant for the first time were determined as follows: gymconopin D (10), 3, 5- diimethoxy-3’-hydroxy- bibenzyl(12), Kayaflavone(15), Amentoflavone (16), methyl (4-OH) phenylacetate (17), p-hydroxy benzaldehyde(18), p-hydroxybenzcic acid(19), 4-(methoxymethyl) phenol(20), Methyl 3-(4-hydroxyphenyl)propionate(21), Methyl 3-(3-hydroxyphenyl)propionate(22), cinnamic acid(23), 3-hydroxybenzcic acid(24), 4-(ethoxymethyl) phenol(25), 4-oxopentanoic acid(26), 5-hydroxymethyl ffurfural(27), 3-hydroxyphenylpropionic acid(28),ρ-dihydroxy benzene(29), gastrodine (30)Moreover, PE, EtOAC, n-BuOH extract of P. bulbocodioides and the isolated compounds were screened by cytotoxity experiments. PE, EtOAC extract as well as compounds 4, 7, 12, 13, 29 have some activity to inhibit LA795 cells The studies will be helpful for the further investigaton of the active constituents in this plant..
