一氧化碳吸入对脂多糖诱导急性肺损伤大鼠肺部炎症反应的影响

【摘要】 实验一、一氧化碳吸入对脂多糖诱导急性肺损伤 大鼠动脉氧合功能的影响 目的:观察低浓度一氧化碳(CO)吸入对脂多糖(LPS)诱导急性肺损伤(ALI)大鼠动脉氧合功能及碳氧血红蛋白(COHb)含量的影响。方法:72只雄性SD大鼠随机数字表法均分为正常对照、单纯CO吸入、ALI和ALI+CO吸入四组,阴茎背静脉注射给药。对照组注入生理盐水,单纯CO吸入组注入生理盐水后持续吸入250ppm CO,ALI组注入LPS 5mg/kg体重,ALI+CO吸入组注入LPS5mg/kg体重诱导ALI后持续吸入250 ppm CO,各组注入液体总量相同。在各组观察的1、3和6 h批次(每批次6只)剖腹,自腹主动脉放血处死,留取动脉血分析血气和COHb浓度。结果:四组组间相应时间点及组内各时间点比较,动脉氧分压(PaO2)、动脉氧饱和度(SaO2)均无统计学差异。COHb(%):单纯CO吸入组[(7.2±1.1)、(6.9±1.4)、(7.1±1.7)]、ALI组[(1.3±0.6)、(1.4±0.4)、(1.3±0.5)]及ALI+CO吸入组[(7.0±1.3)、(6.9±1.1)、(7.1±1.6)]显著升高,与正常对照组相应时间点[(0.6±0.4)、(0.5±0.2)、(0.6±0.3)]比较,差异有统计学意义(P均<0.05),组内各时间点比较,差异无统计学意义;单纯CO吸入组、ALI+CO吸入组的COHb显著高于ALI组相应时间点(P均<0.05)。结论:无论是5mg/kg体重LPS静脉注入,还是250 ppm CO吸入均不影响大鼠动脉氧合、增加COHb含量。

【Abstract】 Section 1: Effects of carbon monoxide inhaled on arterial oxygenation of rats with acute lung injury induced by lipopolysaccharideObjective: To observe the effects of carbon monoxide (CO) inhaled on arterial oxygenation and carboxyhemoglobin (COHb) level of rats with acute lung injury (ALI) induced by lipopolysaccharide (LPS). Methods: Seventy-two male Sprague-Dawley rats were randomly divided into four groups. Drugs were injected by intravenous. Control group received normal saline, CO inhaled alone group received an inhalation 250 ppm CO, ALI group received 5 mg.kg-1 LPS, ALI+CO inhaled group received an inhalation of 250 ppm CO after ALI induced by 5 mg.kg-1 LPS injected. The animals were sacrificed by exsanguinations at 1,3 and 6 h of the observation in four groups. Arterial blood was harvested for determinating the partial pressure of arterial oxygen (PaO2), saturation of arterial oxygen (SaO2) and COHb level. Results: PaO2 and SaO2 of four groups had statistics difference in neither the same time point of groups nor the differential time point of the same group. COHb of CO inhaled alone group and ALI+CO inhaled group was significantly increased compared with control group at the same time point (all P<0.05).
