内皮素-11-31的心血管效应及其机制研究

【摘要】 ET-11-31是糜酶水解大内皮素产生的由31个氨基酸组成的内皮素家族的新成员。ET-11-31可直接激活ET受体,也可由ECE水解为ET-11-21间接产生效应。在外周,ET-11-31有收缩平滑肌、促进细胞增殖等生物学作用。ET-11-31的心血管效应尚未明确。本工作证明经冠状动脉口给予ET-11-31产生升压作用,无降压作用。中枢给予ET-11-31产生与ET-11-21类似的心血管效应,ET-11-31的中枢心血管效应通过转换为ET-11-21间接产生,不是ET-11-31本身作用。ET-11-31有增高培养心肌细胞[ca2+]i的作用,10-8mol/ET-11-31能减小钙瞬变幅度,该作用部分是ET-11-31的直接作用。ET-11-31的中枢心血管效应和增加胞浆[Ca2+]的作用是由ETA受体介导,与ETB受体关系不大。

【Abstract】 Abstract: ET-11-31, a novel member of the endothelin family comprising 31 amino acids and derived from the selective hydrolysis of big ET-1 by chymase. Although the peripheral effects of ET--11-31 have been well determined, the cardiovascular role of ET--11-31 is not fully understood. Our study suggested that administration of ET--11-31 through coronary artery increased blood pressure of rats. Central administration of ET--11-31 produced biphasic cardiovascular effects. The central effects of ET--11-31 which were mediated through ETA receptors and did nothing with ETB receptors, result from ET--11-31 converting to ET--11-21 ET--11-31 increase [Ca2+]I, but 10-8 mol/L terminated [Ca2+]I, which were partly attenuated by phosphoramidon.
