中药方剂四物汤补血作用有效部位的研究

【摘要】 目的在四物汤已有的研究结果基础上,确定四物汤全方的补血有效部位。对“有效部位”建立完整的质量控制体系,并进行补血作用机理研究。方法立足于四物汤全方,将全方的成分分离为三个大的有关部位。研究过程将中药化学、中药质量控制以及药理学学科紧密结合。通过血清药理学、血清药化学和药代动力学实验,对四物汤补血作用机理进行研究。成果完成了CO2超临界萃取部位、醇提部位以及多糖部位的分离、分析;建立了与四物汤治疗血虚证的血象特点相对应的血虚模型;确定了四物汤补血有效部位;对有效部位建立了完整的质量控制体系,并进行了指纹图谱研究;对有效部位开展了血清药化学、血清药理学和药代动力学研究。结论本文确定了四物汤的补血有效部位,其化学成分主要包括多糖、苷类、有机酸类和生物碱类。有效部位的质量标准为确保用药的有效性提供了依据。血清药化学实验确定了阿魏酸是有效成分之一。血清药理学研究结果表明,四物汤及其有效部位均可以显著促进骨髓细胞的分化和增殖。药代动力学参数为临床用药提供了依据。

【Abstract】 Objective On the bases of investigation data , the paper had fixed the effective fraction of SWT about enriching blood action. Set up complete quality control system for the effective fraction. Carried out the studies on the mechanism of enriching blood. Methods The investigation was based on full recipe of SWT and separated all chemical components into three parts. During the sduty , Chinese traditional medicine chemistry, quality control as well as pharmacology was combined together closely. Result The paper had separated SFE-CO2 position, alcohol position as well as polysaccharides position, then analyzing the chemical component within them. Blood deficiency model according with hemogram characteristic of clinic "pattern" was set up , then the effective fraction of SWT was fixed. Set up complete quality control system for the effective fraction and carried out the research of Fingerprints. Carried out the studies on the mechanism of enriching blood by Serum pharmacology, Serum chemistry and pharmacodynamics test. Conclusion The paper determined the effective fraction of enriching blood about SWT which included polysaccharides. glycosides, organic acids and alkaloids.The quality standard of effective fraction provided basis for the validity of drug. Ferulic acid was defined as one of effective component by Serum chemistry. The result of Serum pharmacology showed that SWT and its effective fraction all can promote hyperplasia and differentiation of marrow cell .Study on pharmacokinetics provided parameters for clinic.
