炎克宁对大鼠输卵管阻塞性不孕模型作用机理的研究

【摘要】 为了揭示炎克宁治疗输卵管炎性阻塞性不孕的作用机制,以Wistar雌性大鼠为受试对象,用混合菌接种法复制输卵管炎性阻塞性不孕模型,96只大鼠随机分为正常对照组、模型组、中药对照组、炎克宁低剂量组、中剂量组、高剂量组,每组又分为15天、30天两个时间组。采用HE染色观察输卵管组织形态学变化、电镜观察输卵管组织超微结构、原位末端标记(TUNEL染色)法检测输卵管上皮细胞凋亡、应用免疫组化和原位杂交技术检测输卵管上皮细胞BcL-2、EGFR和ICAM-1蛋白和mRNA表达,ELISA法检测血清中IL-1、IL-6和TNF-a含量。 实验结果显示:①炎克宁高剂量组与模型组、中药对照组、炎克宁低剂量组、炎克宁中剂量组比较,能明显抑制输卵管细胞凋亡,减轻输卵管粘膜充血、水肿、炎性渗出、炎性粘连,促进组织修复;②炎克宁高剂量组输卵管上皮细胞Bcl-2蛋白和mRNA表达高于模型组、中药对照组、炎克宁低剂量组、炎克宁中剂量组;③炎克宁高剂量组ICAM-1蛋白和mRNA表达在输卵管上皮细胞的表达明显低于与模型组、中药对照组、炎克宁低剂量组、炎克宁中剂量组;④炎克宁高剂量组输卵管上皮EGFR蛋白和mRNA表达低于模型组、中药对照组、炎克宁低剂量组、炎克宁中剂量组;⑤炎克宁高剂量组血清中IL-1、IL-6和TNF-a明显低于模型组、中药对照组、炎克宁低剂量组、炎克宁中剂量组。 结论:①用混合菌接种法可以复制大鼠输卵管炎性阻塞性不孕模型;②炎克宁能明显抑制输卵管细胞凋亡,减轻输卵管粘膜充血、水肿、炎性渗出、炎性粘连,促进组织修复;③炎克宁促进Bcl-2蛋白和mRNA表达很可能是其抗凋亡的机制之一;④炎克宁能够抑制ICAM-1蛋白和mRNA表达,从而发挥其抗炎症作用;⑤抑制输卵管上皮EGFR蛋白和mRNA过度表达很可能是炎黑龙江中医药大学博士学位论文克宁治疗输卵管炎性阻塞性不孕途径之一;⑥炎克宁治疗输卵管炎性阻塞性不孕可能与降低血清中IL一1、IL一6和TNF一a含量有关.

【Abstract】 To study on the mechanism of Yankening treating fallopian tube inflammatory obstructive infertility(FTIOI),the 96 rats were inoculated mixed bacterium to copy the model, which were divided into the normal group,the model group, the TCM controlled group,the lower dosage group, the middle dosage group, the high dosage group, while the every group was divded into two time group which were 15 days and 30 days.HE staining was used to observe the change of fallopian tube in morpholog ,while to observe the ultrastructure with electron microscope,to detect the cell apoptosis with TUNEL staining,to detect BCL-2,EGFR and ICAM-lmRNA with immunohistochemistry and hybridization in situ, and to detect IL-l,IL-6,TNF-ain serum with ELISA.The results showed that: (1) To compared with the model group, the TCM controlled group,the lower dosage group, the middle dosage group, the high dosage group can restrain the cell apoptosis,relieve hyperemia, edema, adherence and granuloma,and accelerate tissue repair; (2)The high dosage group can improve the expression of BCL-2 mRNA and protein;(3)The high dosage group can decrease the expression of ICAM-lmRNA and protein;(4)The high dosage group can decrease the expression of EGFR mRNA and protein; (5)The high dosage group can decrease the contentof IL-l,IL-6 and TNF-ain serum.The conclutions: (1)Mixed bacterium inoculated can copy the model; (2)The middle dosage group, the high dosage group can restrain the cell apoptosis,relieve hyperemia, edema, adherence and granuloma,and accelerate tissue repair;(3)By improving theexpression of BCL-2 mRNA and protein,the Yankening can anti-apoptosis;(4)By decreasing the expression of EGFR mRNA and protein, Yankening can have anti-inflammatory action; (5)The high dosage group can decrease the contentof IL-l,IL-6 and TNF-ain serum.
