夹脊电针对脊髓损伤大鼠治疗作用的研究

【摘要】 本课题以Wistar大鼠经Allen’s法制备脊髓撞击伤模型,实验分成两部分: 实验一:对相同损伤节段采取不同时间进行夹脊电针治疗,即伤后8h内和伤后24h接受夹脊电针治疗或地塞米松治疗,以联合行为评分法测定不同时间模型大鼠的运动功能、感觉功能,以免疫组化法及光镜技术、观察各不同时间点伤段脊髓的NT-3及c-Fos表达,结果表明:伤后8h内进行治疗较伤后24h进行治疗,能更好地促进模型大鼠的运动及感觉功能恢复,而伤后8h内进行治疗的近期疗效,地塞米松的治疗效果好于夹脊电针的治疗效果,但中远期疗效,夹脊电针的治疗效果优于地塞米松的治疗效果;伤后24h治疗,地塞米松无优势,其夹脊电针的中远期效果较好。 实验二:对不同损伤节段在相同时间进行夹脊电针治疗及夹脊电针加中药脑髓再生丹治疗,以原位杂交及光镜技术检测伤段脊髓内源性神经营养因子(NGFmRNA、BDNFmRNA),原癌基因c-FosmRNA的表达,结果表明:不同损伤节段通过针刺及中药治疗,其NGFmRNA、BDNFmRNA及c-FosmRNA的表达时间不同,经治疗后T8节段较L1节段神经营养因子及原癌基因的表达时间早。 本实验从分子水平阐述了夹脊电针治疗脊髓损伤的积极作用,能够促进内源性神经营养因子分泌、促进神经再生。证实了夹脊电针治疗脊髓损伤存在时间窗口。同时证实了夹脊电针治疗脊髓损伤有空间差别。本实验为临床治疗脊髓损伤提供了理论和实验基础。

【Abstract】 In this research, taking Wistar rats models of SCI made by Allen’s as objects of study. There are two part in this research.Research one: in the same segments, treatment are taken by Jiaji electropuncture or dexamethasone (DXM) in 8h and 24h after SCI, model rat’s motor and sensory function are studied by CBS; injury spinal cord’s NT-3 and c-Fos are studied by ABC immunohistochemistry and optic techniques. The result show that model rats’ motor and Sensory function treatment are apllied in 8h is better in 24h, short time effect, DXM effect is better than Jiaji electropuncture, remote effect, Jiaji electropuncture is better than DXM when treatment are applied in 8h; remote effect, DXM have no ascendancy, but Jiaji electropuncture effect is good when treat ment are applied in 24h.Reseach two: in the different segments, treatment applied by Jiaji electropuncture and NaosuizaishengDan or Jiaji electropuncture at same time, with advanced techniques such as insitu hybridization and optic, endogenous NTFS. c-Fos are studied. The result show that: The NGFmRNA, BDNFmRNA, c-FosmRNA’s, postive incidence time is different at different segments, the highest level time of each factor are earlier in T8 than in L1.This article elaborates the active influnce of Jiaji electro-puncture to treatment SCI at molecular level. Jigji electropuncture can enhance endogene NTFS secret and promote nerve the regenration. It verify that there is new treatmant time and space difference treatment appling by Jiaji electropuncture to SCI.The research provides theoretical and experimental foundation for clinical treatment to SCI.
