电针刺激夹脊穴对大鼠后肢血管舒张作用的实验研究

【摘要】 目的:1、观察电针刺激大鼠背部L3-5夹脊穴对大鼠后肢皮肤血流的影响。2、观察注射辣椒素类似物(RTX)、降钙素基因相关肽8-37、ATP敏感性钾离子通道拮抗体格列本脲、钙离子敏感性钾离子通道的拮抗体蝎毒素等试剂对电针刺激大鼠夹脊穴皮肤血流改变的影响,探讨电针夹脊穴与脊髓刺激诱导肢体血管舒张的相关性。3、通过对累及刺激夹脊穴后大鼠血浆中的NO、ET-1、6-K-PGF1α、TXB2含量改变,以及动脉壁eNOS、ET-1表达水平的观察,探讨电针夹脊穴舒张大鼠后肢血管的作用机理。方法:1、将清醒Wistar大鼠固定,针刺大鼠背部L3-5夹脊穴,然后接电针刺激,比较各阶段大鼠皮肤血流PU值的改变。2、电针刺激大鼠夹脊穴后,分别注射辣椒素类似物(RTX)、降钙素基因相关肽8-37、ATP敏感性钾离子通道拮抗体格列本脲、钙离子敏感性钾离子通道的拮抗体蝎毒素,比较各试剂注射前后刺激大鼠夹脊穴后肢皮肤血流PU值的改变。3、大鼠随机分为空白组与电针组,电针组每日电针大鼠背部L3-5夹脊穴,累计刺激15日后两组大鼠采血,ELISA法检测NO、ET-1、6-K-PGF1α、TXB2含量。4、将大鼠随机分为空白组、短期刺激组、长期刺激组。短期组与长期组大鼠每日电针背部L3-5夹脊穴,短期组累计刺激7日,长期组累计刺激15日。分别取三组大鼠股动脉,用Western Blot检测大鼠动脉壁eNOS、ET-1表达水平。结果:1、电针刺激大鼠L3-5夹脊穴可增加大鼠后肢皮肤血流PU值。2、辣椒素类似物(RTX)、降钙素基因相关肽8-37、格列本脲、蝎毒素可抑制电针夹脊穴诱导的皮肤血流增加。3、经过累计刺激后大鼠血浆中NO、6-K-PGF1α含量升高,ET-1、TXB2含量下降,后肢动脉eNOS表达增强,ET-1表达下降。结论:电针刺激大鼠L3-5夹脊穴可以诱导大鼠后肢血管舒张,而且这种舒张效应与脊髓刺激的结果类似。同时累计刺激可以改变大鼠血浆中舒缩血管物质的含量,并调节内皮细胞功能。为临床治疗PAOD疾病提供了新的治疗方法。

【Abstract】 Objective:1. To observe the effects of electro-acupuncture dorsal L3~5 Jiaji points on skin blood flow in hind limbs of rats.2. To observe the effects of injecting capsaicin analogues (RTX), calcitonin gene related peptide 8-37, ATP sensitive K- channel antagonist glibenclamide, Ca2+ sensitive K+ channel antagonist scorpion toxins on skin blood flow during electro-acupuncturing Jiaji points of rats, and to investigate the relationship between electro-acupuncturing Jiaji points and the limb vasodilation by spinal cord stimulation.3. To discuss the mechanism of accumulatively electro-acupuncturing Jiaji points on vasodilation of rats hind limbs by counting the content changes of NO, ET-1,6-K-PGF1αand TXB2 in plasma, as well as the expression of eNOS and ET-1 in arterial wall.Methods:1. Conscious Wistar rats were fixed and stimulated on Jiaji points by general acupuncture and electro-acupuncture, respectively, and then the PU values of skin blood flow at different stages were compared.2. After electro-acupuncture stimulation on Jiaji points, the rats were injected capsaicin analogues (RTX), calcitonin gene related peptide 8-37, ATP sensitive K+ channel antagonist glibenclamide, Ca2+ sensitive K+ channel antagonist scorpion toxins, respectively, and then the PU values of skin blood flow before and after the injection were compared.3. The rats were randomly divided into two groups, blank group and electro-acupuncture group, and the electro-acupuncture group was daily stimulated on dorsal L3~5 Jiaji points for fifteen days. After that, the ELISA method was used to check the content of NO, ET-1,6-K-PGF1αand TXB2 of two groups.4. The rats were randomly divided into two groups, blank group, short-term electro-acupuncture group and long-term electro-acupuncture group, and the latter two groups were daily stimulated on dorsal L3~5 Jiaji points for seven and fifteen days, respectively. After that, the femoral arteries of three groups were taken and analyzed on the expression of eNOS and ET-1 by Western Blot technique. Results; 1. Electro-acupuncturing dorsal L3~5 Jiaji points can enhance the PU values of skin blood flow in hind limbs of rats.2. The enhancement of skin blood flow induced by electro-acupuncture can be inhabited by the injection of capsaicin analogues (RTX), calcitonin gene related peptide 8-37, glibenclamide, scorpion toxins.3. After accumulative stimulation, the contents of NO and 6-K-PGF1αin rats plasma were increased, while those of ET-1 and TXB2 were decreased; the expression of eNOS in hind limbs artery was enhanced, while that of ET-1 was weakened.Conclusions:Electro-acupuncturing dorsal L3~5 Jiaji points can enhance the vasodilation of rats hind limbs, which is quite similar to the result induced by spinal cord stimulation. Accumulative electro-acupuncture stimulation can change the contents of some vasoconstrictor and vasodilator, and can adjust endothelial cell function. Electro-acupuncture will be a novel treatment for PAOD disease in clinical practice.
