氢水对急性马尾综合征大鼠神经元损害抑制作用的实验研究

【摘要】 目的:研究氢生理盐水在急性马尾综合征大鼠中的抗氧化应激反应、抑制神经元凋亡及神经保护作用。方法:将大鼠随机分为5组:正常组、压迫组、氢水组、甲强龙组、氢水和甲强龙混合治疗组,在不同的时间段进行BBB评分和热板实验行为学观察、MDA和GSH水平检测、行组织HE染色,NF染色,Caspases 3、Bax及Tunel染色,对动物行为学、氧应激反应程度、组织形态学改变、细胞凋亡情况进行综合评估。结果: (1)压迫组与正常组相比,行为学、组织形态学、氧化应激指标和细胞凋亡在48小时内具有明显差异;(2)氢水组与压迫组相比,BBB评分和热板实验在6至48小时内具有较大差异,48小时至12周无明显变化;(3)与压迫组相比,氢水组脊髓、马尾及背根神经节的MDA水平更低,GSH水平更高;压迫组3小时MDA和GSH变化幅度最大,氢水组24h MDA和GSH变化幅度最大;(4)与压迫组相比,氢水组HE染色显示出更好的组织形态,马尾NF阳性率更高,而脊髓及背根神经节的Caspases 3和Bax阳性指数、Tunel细胞凋亡指数均更低;(5)氢水组、甲强龙组、氢水和甲强龙混合治疗组之间无明显的统计学差异,与压迫组有明显差异。结论:急性马尾综合征大鼠的氧化应激反应在3小时迅速增强,48小时达到峰值,氢生理盐水对氧化应激反应具有抑制作用;氢生理盐水对神经元凋亡具有抑制作用,对神经损害具有保护作用;与甲强龙相比,氢生理盐水具有类似疗效。

【Abstract】 Objective To study the antioxidational, antiapoptotic,and neuroprotective effects of hydrogen-rich saline in rats with acute cauda equina syndrome. Method Rats were randomly allocated into five groups: normal group, compression group,hydrogen-rich saline group, methylprednisolone group, and methylprednisolone combined hydrogen-rich saline group. In different periods, they respectively were examined by BBB score and the hot plate test, their samples were harvested to analyze tissue concentrations of MDA and GSH, and perform HE, NF, Caspases 3, Bax and Tunel staining. By above methods, animal ethology, oxidative stress, histomorphology and cell apoptosis were evaluated. Result (1) Compared with the normal groups, compression groups had significant differences of histomorphology, cell apoptosis, animal ethology and oxidative stress, especially in 48 hours. (2)Compared with the compression groups, hydrogen-rich saline groups had significant statistical differences of BBB scale and hot plate test in 6 to 48 hours groups, that were opposite in 48 hours to 12 weeks groups. (3) Compared with the compression groups, hydrogen-rich saline groups had lower MDA level and higher GSH level which had statistical significance in spinal cord, cauda equina and dorsal root ganglia; MDA and GSH level noticeable changed in 3 hours compression groups and 24 hours hydrogen-rich saline groups. (4)Compared with the compression groups, hydrogen-rich saline groups showed better morphology of HE staining, higher positive rate of NF staining of cauda equina, lower positive index of Caspases 3 and Bax staining, and lower Tunel apoptotic index of the spinal cord and dorsal root ganglia, above data all had statistical significant. (5) Compared with the compression groups, experimental data of methylprednisolone, hydrogen-rich saline and methylprednisolone combined hydrogen-rich saline groups all had statistical differences, while there weren’t statistical differences between methylprednisolone, hydrogen-rich saline and methylprednisolone combined hydrogen-rich saline groups. Conclusion Oxidative stress of acute cauda equina syndrome in rats appeared the largest rate of change at 3 hours and reached peak at 48 hours. Hydrogen-rich saline had antiapoptotic and neuroprotective effects in acute cauda equina syndrome. Compared with methylprednisolone, hydrogen-rich saline had a similar therapeutic effect.
