慢性时差模型的情绪时间动力学特征及其针刺干预机制研究

【摘要】 目的:研究慢性时差对金黄地鼠的情绪时间动力学特征的影响,探讨针刺对慢性时差模型情绪紊乱的时间动力学特征的调节效应及其作用机制。方法:运用超前性光周期转移方法,复制慢性时差金黄地鼠模型。(1)采用高架十字迷宫和旷场实验观察模型动物6个时间点ZT0(8:00)、ZT4(12:00)、ZT8(16:00)、ZT12(20:00)、ZT16(24:00)、ZT20(4:00)的情绪变化;(2)造模结束后,选用ZT4电针“百会”、“长强”,电压1-3v,频率15-25Hz,时间15分钟,针刺3天,1次/天,观察针刺对动物的情绪时间动力学特征的影响。(2)应用实时荧光定量PCR法检测SCN内AVP基因表达、下丘脑区AVP和CRH基因表达,采用放射性免疫分析法检测垂体AVP水平,运用酶联免疫法检测血清CORT含量,探讨电针对慢性时差模型情绪时间动力学特征影响的作用机理。结果:(1)模型组动物焦虑情绪和抑郁情绪总体水平高于空白组。两组均未显现明显的昼夜节律特性,但正常动物焦虑和抑郁情绪呈现双峰双谷的时间动力学模式(似超日节律);造模后表现双峰双谷特征消失,或有双峰但峰值相位移相或反相,中值降低、振幅过大或过小等异常时间动力学特征。OT%、OE%两项指标不同测量时间差异有统计学意义,即总体水平有随时间变化趋势,分组因素有作用,时间因素因分组不同而不同。水平移动格数、垂直运动次数也存在时间因素因分组不同而不同。(2)自然恢复组动物焦虑、抑郁情绪双峰双谷特征结构消失,表现单峰形式、或出现昼夜节律。(3)电针治疗后动物情绪多项指标双峰双谷的时间动力学特征结构以及中值、振幅等节律参数有所恢复。(4)与空白组比较,模型组动物SCN内AVP基因表达、垂体AVP水平有明显降低、血清CORT有明显增高,自然恢复组SCN内AVP基因表达有明显下降、血清CORT有明显增高;针刺组较模型组动物SCN内AVP基因表达和垂体AVP水平显著升高,较自然恢复组SCN内AVP基因表达有显著增高、血清CORT有降低趋势。结论:正常金黄地鼠情绪呈现双峰双谷的时间动力学结构模式,慢性时差后动物情绪时间动力学特征异常(反相、移相或形成单峰),针刺治疗可在一定程度上恢复动物情绪的双峰双谷的时间动力学结构,其作用机制可能与电针能上调中枢AVP的水平和基因表达、抑制血清CORT异常增高,从而调节HPA轴的功能相关。

【Abstract】 Objective:The aim of this study was to investigate the Effect and Mechanism of Electro-acupuncture (EA) on Characteristics of Temporal Dynamics of the Mood in Syrian hamster Model of Chronic Jet Lag.Methods:Model of chronic jet lag was established by appling the methods of advances of light/dark cycle for 10 days. EA (15/25Hz in frequency, 1-3v) was applied to "Baihui" (Du20) and "Changqiang" (Dul) for 15min/d and lasted for 3 days. The behavioral tests were measured by the elevated plus-maze test (EPM) and open field test (OF) in ZTO(8:00)、ZT4(12:00)、ZT8(16:00)、ZT12 (20:00)、ZT16 (24:00)、ZT20 (4:00).The expressions of AVPmRNA and CRHmRNA were measured by PR-PCR method. The level of AVP in the pituitary gland was measured by in radio-immunity. The content of CORT was detected by ELISA method.Results:(1) Compared with the control group, the levels of anxiety and depression were increased in model group. Double-peak and double-valley characteristics of temporal dynamics of the mood (similar ultradian rhythms) could be observed in control group.Abnormal characteristics of temporal dynamics of the mood were represented in the model group (eg.phase shift,antiphase of the peak performance phase, the midvalues decrease, and the amplitude large or small). (2) Abnormal characteristics of temporal dynamics of the mood was represented in the Natural recovery group (eg.Circadian rhythm with single peak). (3) Double-peak and double-valley characteristics of temporal dynamics of the mood (similar ultradian rhythms) was appeared in EA group.(4) Compared with the model group and the Natural recovery group, AVPmRNA in the SCN and AVP in the pituitary were were obviously increased, AVPmRNA in the hypothalamus and CORT in serum were displayed a recovery trend by EA.Conclusion:Double-peak and double-valley characteristics of temporal dynamics of the mood was showed in control group. Abnormal characteristics of temporal dynamics of the mood was displayed in the model group and the natural recovery group(eg.disappearance and phase shift of Double-peak and double-valley, or single peak). Double-peak and double-valley temporal dynamic structure of the mood can be restored by EA. The mechanism of EA may be correlated with affection AVP level and gene expression in the center, regulating HPA axis function.
