白术茯苓汤调控脾虚大鼠血管活性肠肽的物质基础研究

【摘要】 目的:研究白术茯苓汤调控脾气虚大鼠血管活性肠肽的化学成分或组分,并探讨各化学成分或组分的配伍协同作用。方法:①采用系统溶剂法从白术茯苓汤萃取分离得到其有效部位,进一步运用现代中药化学的植化分离手段,对有效部位进行分离鉴定得到各组分。②运用中药色谱技术研究白术茯苓水煎液中白术内酯Ⅲ的来源以及白术、茯苓不同配比对其含量的影响。③复制脾气虚证的动物模型,观察白术茯苓汤各提取分离组分及组分配伍对脾气虚大鼠血管活性肠肽、血管活性肠肽受体及受体结合率的影响,确定有效组分。结果:①1从白术茯苓汤水煎液提取分离出白术茯苓总多糖、总络合物、白术内酯Ⅲ。②高效液相色谱结果显示,白术茯苓汤复方水煎液出峰的强度和个数与白术相似,而与茯苓差异较大;在215nm条件下,白术茯苓汤复方水煎液出峰的强度最大,峰的个数最多;白术茯苓汤水煎液中,白术:茯苓复方按照1:1、3:1、2:1、1:3、2:3、3:2、1:2的变化,白术内酯Ⅲ含量发生了不同程度的依次降低。③与模型组比较,白术茯苓总多糖高剂量组、白术内酯Ⅲ高剂量组、络合物高剂量组、组分配伍高剂量组、白术茯苓汤组血浆血管活性肠肽降低(P<0.05及P<0.01),白术内酯Ⅲ高剂量组、络合物高剂量组、组分配伍高剂量组、组分配伍中剂量组、白术茯苓汤组肠组织血管活性肠肽降低(P<0.05)与模型组比较,白术茯苓总多糖高剂量组、白术内酯Ⅲ中剂量组、络合物高剂量组、组分配伍中剂量组、白术茯苓汤组血管活性肠肽受体1(VIPR1)降低,白术内酯Ⅲ高剂量组、络合物高剂量组、白术茯苓总多糖高剂量组、络合物中剂量组、组分配伍中剂量组、白术茯苓汤组血管活性肠肽受体1(VIPR2)降低(P<0.05及P<0.01);与模型组比较,上述三组分配伍之高剂量组和白术茯苓汤组能降低脾气虚大鼠受体结合率水平。(P<0.05)结论:①白术茯苓汤下调脾气虚大鼠血管活性肠肽及血管活性肠肽受体的物质基础为白术茯苓总多糖、总络合物、白术内酯Ⅲ。②白术茯苓汤复方水煎液的成分主要来自白术,白术内酯Ⅲ是白术茯苓汤的重要化学成分之一。③白术茯苓汤中,白术与茯苓不同的配比对白术内酯Ⅲ的含量有影响,其中白术:茯苓(1:1)复方中白术内酯Ⅲ的含量最高,白术:茯苓(1:2)复方中白术内酯Ⅲ的含量最低。

【Abstract】 Objective:To research the chemical compositions or components extracted from Baizhufuling Tang that can regulate Vasoactive Intestinal Peptide (VIP) and its receptor on the Spleen—Qi deficient rat, study the enhancement effect owing to compatibility and synergy of the chemical ingredients or components.Methods:①By using the system solvent extraction from the Bai Zhu Fu Ling Tang we isolated active component, and by further using modern Chinese chemical plant chemical means for isolation and identification, we obtained the effective part of all the components.②By applying chromatographic technique we rsearched the source of atractylenolide III in the decoction of Bai Zhu Fu Ling Tang and the influence of Atractylodes, Poria different proportion to the content of atractylenolideⅢ.③By Using the rat model of Spleen-Qi deficiency by the method of relieving stagnation of Qi and bitter cold starve and full in abnormal, we researched the influence of different components extracted from Baizhufuling Tang and combinations of these component on the Spleen Qi deficient rat, compared the different groups effect on the VIP, VIP receptor and receptor-binding of the rats, thence determined the effective components.Result:①Baizhufuling total polysaecharide, total complex compounds and atractylenolide III were isolated and extracted from Bai Zhu Fu Ling Tang decoction.②It is shown that Bai Zhu Fu Ling Tang decoction is similar to Atractylodes Decoction in the intensity and number of chromatographic peak, is largely different from Poria Under the condition of 215nm wavelength, the intensity of chromatographic peak of Bai Zhu Fu Ling Tang decoction is largest, and the number of chromatographic peak is most. In Bai Zhu Fu Ling Tang decoction, the content of atractylenolide③decreases in proper sequence when the proportion of Atractylodes and Poria change as the sequence of 1:1,3:1,2:1,1:3,2:3,3:2 and 1:2.③Compared with model group, VIP in blood plasma of the high dosage of Baizhufuling total polysaecharide group, the high dosage total complex compounds group, the high dosage atractylenolideⅢgroup and the high dosage of compound synergy group, Baizhufuling group was reduced (P<0.05 or P<0.01), Compared with model group; VIP in colon tissue of the high dosage of total polysaecharide group, the high dosage atractylenolide III group and the high dosage of compound synergy group, the middle dosage of compound synergy group, Baizhufuling group was reduced (P<0.05) Compared with model group, vasoactive intestinal peptide receptor 1 (VIPR1) in colon tissue of the high dosage of Baizhufuling total polysaecharide group, the middle dosage atractylenolideⅢgroup and the middle dosage of compound synergy group, Baizhufuling group was reduced (P<0.05 or P<0.01), Compared with model group, vasoactive intestinal peptide receptor 1 (VIPR2) in colon tissue of the high dosage of Baizhufuling total polysaecharide group, the high dosage of atractylenolideⅢgroup, the high dosage of compound synergy group, and the middle dosage of compound synergy group, Baizhufuling group was reduced (P<0.05 or P<0.01); Compared with model group, VIP receptor-binding in colon tissue of the high dosage of compound synergy group and Baizhufuling group was reduced (P<0.05)Conclusion:①Baizhufuling total amylose, total complex compounds and atractylenolideⅢare the Material Basis of Bai Zhu Fu Ling Tang that lower VIP and VIP receptor of the Spleen Qi deficient rat.②Atractylodes is the main source of the components of Bai Zhu Fu Ling Tang decoction and atractylenolide III is one of the important ingredients.③In Bai Zhu Fu Ling Tang decoction, the content of atractylenolideⅢis influenced by the the proportion of Atractylodes and Poria, and it is highest when the proportion of Atractylodes and Poria is 1:1, is lowest when the proportion of Atractylodes and Poria is 1:2.
