欧前胡素逆转A2780/Taxol多药耐药作用及其机制研究

【摘要】 目的:探讨欧前胡素对人卵巢癌耐紫杉醇细胞株A2780/Taxol耐药性的逆转作用及其可能的作用机制。方法:采用四甲基偶氮唑盐(MTT)比色法检测欧前胡素对人卵巢癌细胞A2780和人卵巢癌耐紫杉醇细胞株A2780/Taxol增殖的影响;观察欧前胡素作用A2780/Taxol细胞24h之后对紫杉醇耐药性的逆转效果;采用高效液相-荧光检测法和Western blot分别对其机制进行研究。结果:A2780/Taxol对紫杉醇耐药倍数为4.34(1.5<RI<5)倍;不同浓度欧前胡素对Taxol的逆转倍数分别为2.89、3.38、6.76,且呈剂量依赖性增加。不同浓度的维拉帕米及欧前胡素可显著增加A2780/Taxol细胞株内Rho123的蓄积。不同浓度的欧前胡素能够显著下调A2780/Taxol细胞株的P-gp表达量。结论:欧前胡素能够部分逆转A2780/Taxol细胞的耐药性,且其逆转机制可能与降低P-gp的表达量相关。

【Abstract】 Objective: To study the reverse effect and mechanism of imperatorin on the drug resistance of A2780/Taxol cells. Methods: MTT was used to study its effect on proliferation of A2780 and A2780/Taxol cell lines. Drug resistance of A2780/Taxol cell lines against paclitaxel was observed after treated by imperatorin for 24 hours. HPLC-FLD and western blot were used to study its mechanism. Results: The results showed that the multidrug resistance of A2780/Taxol against paclitaxel was 4.34 times(1.5<RI<5). The reversal multiples of different doses of imperatorin against A2780/Taxol were 2.75, 3.47 and 6.29 respectively, in a dose-dependent manner. The accumulation of Rho123 in A2780/Taxol cell lines was significantly increased by different concentrations of imperatorin and Verapamil. The expression of P-gp was reduced by different concentrations of imperatorin. Conclusion: Imperatorin could partly reverse the resistance of A2780/Taxol and its mechanism might be related to the reduction of P-gp expression.
