RCAS1在肾透明细胞癌中的表达研究

【摘要】 【目的】探讨RCAS1/EBAG9在肾透明细胞癌(RCC)中的表达及临床意义。【方法】应用免疫组织化学法检测RCAS1在62例RCC组织及14例癌旁肾组织中的表达,并用Western blot和定量PCR分别检测了6例肾组织(3例肾透明细胞癌和3例癌旁肾组织)中RCAS1的蛋白和mRNA表达。【结果】免疫组织化学结果发现在RCC组织中RCAS1表达率为79.0%(49/62),与癌旁组织表达率14.3%(2/14)相比差异有统计学意义(P<0.01)。RCAS1在RCC的表达随着病理分级增加而增强(P<0.05),与RCC的病理分化程度呈负相关(P<0.01),与RCC的临床分期呈正相关(P<0.01)。Western blot和定量PCR检测RCAS1蛋白和mRNA表达结果显示,RCC组织中的RCAS1表达明显高于癌旁组织(P<0.01)。【结论】RCAS1在RCC中高表达,且与其病理分级、临床分期相关,检测RCAS1的表达有助于对RCC的病理分级、临床分期和预后的判断。更多还原

【Abstract】 【Objective】To investigate the expression of RCAS1 in renal cell carcinoma(RCC) and to study their significance in the progression and prognosis of the RCC.【Methods】Expression of RCAS1 was detected by immunohistochemistry technique in 62 cases of RCC and 14 cases of pare-cancerous renal tissue. And the expression of RCAS1 was also investigated by western blot and real time PCR in 6 cases with renal tissues (3 cases of RCC and 3 cases of pare-cancerous tissue).【Results】The ratio of RCAS1 protein expression was increased in RCC tissues (49/62) compared with pare-cancerous tissue (14.3%, 2/14) (P<0.01) using streptavidin-biotin pemxidase(S-P) method.Within the specimens of RCC group, the differences were statistically significant among WHO pathological grade and clinical stage(P<0.05).There was a negative correlation between the RCAS1 expression and pathological grades(P<0.01),however, there was a positive correlation between the expression of RCAS1 and clinical grades(P<0.01). Both western blot and real time PCR assessed that the expression of RCAS1 in RCC tissues was high than pare-cancerous tissues(P<0.01)【Conclusion】It suggested that high expression of RCAS1 in RCC tissues, and its expression was related to the tumor clinical stage and tumor pathological grade. Detection of RCAS1 expression in RCC was useful to study the pathological grade, clinical stage and prognosis.更多还原
