NADPH氧化酶在高糖诱导的H9c2心肌细胞损伤中的作用
Role of NADPH oxidase in high glucose-induced injury in H9c2 cardiac cells
【摘要】 目的探讨还原型烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶在高糖损伤H9c2心肌细胞中的作用。方法不同浓度(5.5、11、22、33、44、55 mmol·L-1)高糖刺激H9c2心肌细胞24 h及33 mmol·L-1浓度高糖刺激H9c2心肌细胞(0、12、24、36、48、72 h),MTT比色法检测细胞存活率;Western blot检测细胞内Bcl-2、Bax以及NADPH氧化酶亚基p22phox、p47phox以及p67phox的表达水平。结果 33 mmol·L-1浓度高糖刺激H9c2心肌细胞24 h后,细胞存活率开始明显下降,Bcl-2表达减少,而Bax表达增加(P<0.05);此外,NADPH氧化酶亚基p22phox、p47phox以及p67phox蛋白表达水平明显上调(P<0.05)。结论心肌细胞内NADPH氧化酶活性升高可能是介导高糖损伤心肌细胞的重要机制。
【Abstract】 Aim To explore the role of nicotinamide adenine dinucleotide phosphate( NADPH) oxidase in high glucose-induced injury in H9c2 cardiac cells.Methods H9c2 cardiac cells were exposed to different concentrations of high glucose( 5.5 mmol·L-1,11mmol·L-1,22 mmol·L-1,33 mmol·L-1,44 mmol·L-1,55 mmol · L-1) for 24 h and different time pints of high glucose( 0 h,12 h,24 h,36 h,48 h,72h).Cell viability was measured by MTT colorimetry,the protein expression of Bcl-2,Bax and NADPH oxidase submits such as p22 phox,p47phoxand p67 phoxwere determined by western blotting.Results H9c2 cardiac cells exposure to high glucose for 24 h showed on decrease in cell survival and the Bcl-2 expression while an increase in the Bax expression( P < 0.05).Moreover,high glucose could markedly up-regulate the activity of NADPH oxidase characterized by the enhanced expression of p22 phox,p47phoxand p67phox( P < 0.05).Conclusion Activating NADPH oxidase may play an important role in high glucose-induced injury in H9c2 cells.
