复方甘草酸苷对乙型肝炎模型大鼠血清中肝炎相关因子的影响

【摘要】 目的探讨复方甘草酸苷对乙型肝炎模型大鼠血清肝炎相关因子的影响。方法 60只成年健康雄性大鼠,随机分为6组,正常组、模型组、对照组、低、中、高3个剂量实验组,每组10只。正常组与模型组每日灌服等体积的生理盐水3m L,实验组每天灌服3个浓度(10,20,30 mg·kg-1)复方甘草酸苷悬浊液3 m L,对照组组每天灌胃23.3 mg·m L-1拉米夫定溶液3 m L。干预结束后,抽取静脉血3 m L,PCR法检测肝组织中的CD4+T淋巴细胞基因表达,流式细胞法检测T淋巴细胞亚群及激活素a(ACT-a)水平,放免法检测肿瘤因子水平。结果与对照组相比,高、中2个剂量实验组CD4+及CD8+T淋巴细胞亚群水平明显降低,ACT-a及肿瘤坏死因子α(TNF-α)水平也明显降低(均P<0.05)。结论复方甘草酸苷能够明显抑制肿瘤因子的合成,并降低ACT-a的水平,改善CD4+及CD8+T淋巴细胞水平。

【Abstract】 Objective To investigate the effect of the compound glycyrrhizin on hepatitis related factors of hepatitis B in serum of model rat.Methods Sixty specific pathogen free animal rats were selected and divided into 6 groups,normal group,model group,low dose group,middle dose group,high dose group and control group. Normal group and model group were gavaged with the volume of saline 3 m L daily,the experimental group were gavaged with 3 concentrations( 10,20,30mg·kg- 1) of compound glycyrrhizin in 3 m L,the control group were gavaged with concentrations 23. 3 mg·m L- 1of lamivudine solution 3 m L daily. The CD4+T lymphocyte gene expression were detected by polymerase chain reaction method. The T lymphocyte subsets and activin a( ACT- a) level were detected by Flow cytometry. The tumor free factorα( TNF- α) were detected by radioimmunoassay. Results Compared with the control group,CD4+and CD8+Tlymphocyte subsets,TNF- αand ACT- a of the high dose group and middle dose group were significantly lower( P < 0. 05). Conclusion Compound glycyrrhizin can inhibit the synthesis of tumor factor,and reduce the ACT- a levels and improve CD4+and CD8+T lymphocyte level.
