【摘要】 目的观察益肾活血等治法对阿霉素肾病（AN）大鼠的干预作用及与核因子κB（NF-κB）、血栓素A₂（TXA₂）和6-酮前列腺素 F_1α(6-Keto-PGF_1α)等关系,旨在探讨肾虚血瘀证在原发性肾病综合征（PNS）发病中的机制及益肾活血法干预治疗的机理。方法用阿霉素诱导大鼠类似于人类微小病变型肾病（MCD）模型,用泼尼松加肾康灵组治疗,并设泼尼松加知柏地黄汤组和泼尼松加桃红四物汤组为对照组。检测各组治疗前后 NF-κB,血、尿液 TXB₂和6-keto-PGF_1α及肾脏病理形态学等。结果益肾活血中药肾康灵组在降低尿蛋白、提高血清白蛋白（Alb）、降低胆固醇（Ch）和甘油三酯（TG）、减轻肾脏病理损害等作用方面均优于单纯泼尼松组。结论尾静脉注射阿霉素（ADR）造成的人类肾病病理模型,其临床表现和实验指标改变类似于中医的“肾虚血瘀”病变;益肾活血中药肾康灵能显著改善 AN 鼠的肾虚血瘀证表现和实验指标;激素配合益肾活血法中药干预 AN 鼠的疗效在一定程度上强于单纯激素疗法。更多还原

【Abstract】 Objective To observe the intervention function of Yishen Huoxue method on adriamycin-induced nephrotic rats, and its relationship to thromboxane A₂ and prostacychn,with the aim to explore the mechanism of Shen-deficiency and blood stasis syndrome in the occurrence of primary nephropathy syndrome and the mechanism of intervention therapy of Benefiting-Shen and Activating Blood Circulation method.Methods Adriamycin-induced nephrotic rats,which was simi- lar to the nephropathy of minimal changes disease type,were treated with prednisone plus Shenkangling Decoction（a TCM preparation of Shen-benefiting and activating blood circulation,SKL）.Prednisone plus Zhibai Dihuang Decoction and prednisone plus Taohong Siwu Decoction were taken as the control.NF-kB,thromboxane A₂,6-keto-prostaglandin₁ α （6-keto-PGF₁α）and renal pathomorphology were determined in the groups to be tested before and after treatment.Results SKL showed superior effect in lowering urine protein,improving serum Alb,lowering cholesterol and triglyceride,lower- ing NF-kB activity,lessening the renal pathological damage to that in prednisone used only group.Conclusion Patholog- ical model of human nephropathy could be induced by injecting Adriamycin through caudal vein.Its clinical manifestation and experimental indexes were similar to the lesion of“Shen-deficiency and blood stasis”in traditional Chinese medicine. SKL could significantly improve clinical manifestation of Shen-deficiency and blood stasis syndrome and experimental in- dexes.The efficacy got by the principle of benefiting Shen and activating the blood circulation with the intervention of hormone was better than that treated by hormone therapy alone.更多还原
