hApoCⅢ转基因小型猪的生物安全初步评估

【摘要】 为评估hApoCⅢ转基因小型猪的环境生物安全性,采用PCR技术,通过检测其粪便、接触的土壤、肠道菌、代孕母猪的血液DNA以及与之接触的普通猪DNA是否发生了水平基因转移。同时通过大鼠90d经口毒性试验,检测大鼠的血液、生理生化指标和脏器系数是否发生了显著性变化。结果显示:与对照组相比,hApoCⅢ转基因小型猪的肠道内微生物菌群、普通猪的血液和代孕母猪的血液、周围环境土壤都没有发生基因水平转移现象。与对照组相比,试验组大鼠各项观察指标及病理检测均正常,血液、生化指标均在正常范围;初步显示hApoCⅢ转基因小型猪对生物环境是安全的,没有对环境造成不利影响。

【Abstract】 In order to evaluate the biological safety of hApoCⅢ(human apolipoprotein CⅢ)transgenic miniature swine,on the one hand,we would like to know whether it has a genetic drift by testing its feces,contacted soil,intestinal bacteria,and the swine DNA in contact with it and surrogate sow by PCR technology.On the other hand,in 90 days oral toxicity test,rats were measured by the blood and physiological biochemical index and organ coefficient.Results indicated that in comparison with control pig,there was no phenomenon of gene transfer in the microbial flora in the intestinal tract,blood of ordinary pigs and the surrogate sow,and the surrounding soil in hApoCⅢtransgenic miniature pig.Compared with the control,all the indexes and pathological observation of the rats were normal,and the blood and biochemical indexes were in normal range.In conclusion,the data of this study suggested that hApoCⅢ transgenic miniature pigs was safe for the biological environment,no adverse effects on the environment.
