人胚肺二倍体细胞株KMB17在培养过程中的自然凋亡
Natural Apoptosis of Human Embryonic Diploid Lung Fibroblast in Cell Culture
【摘要】 用延长培养周期方法诱导人二倍体胚肺细胞株KMB17自然凋亡,经光学显微镜、荧光显微镜、细胞流式仪、电子显微镜,证明KMB17细胞出现典型的凋亡特征:细胞圆缩、细胞核浓缩破裂、核染色质凝缩后分布于核膜边缘呈新月状和典型的凋亡峰.研究表明人胚肺二倍体细胞(KMB17)在培养过程中易出现自然凋亡.提示可望通过提高细胞对环境压力的耐受能力,预防或延缓细胞培养过程中的自然凋亡,提高操作单元细胞产品的生产效率.
【Abstract】 Cell death in cultures invariably take place by process of apoptosis in response to cell stress.Through natural apoptotisis of human fibroblast (KMB 17) in extended culture.The confluent KMB 17 fibroblast rapidly shrank and disintegrated.by the analysis of light microscope,fluorescent microscope,electronic microscope and flow cytometer,the cell showed apoptotic characteristics of cellular shrinkage,nuclear shrinkage,crescent chromatin around nuclear membrane,even broken nuclear and typical apoptotic peak on flow cytometric figure.In conclusion,the result showed that the natural apoptosis of Human diploid fibroblast (KMB 17) occurred in culture.The result suggests that increasing the antiapoptotic response to cell stress may enhance productivity of cell in culture.
