平阳霉素对TRAIL诱导的胃癌细胞株AGS凋亡的影响
Effect of Pingyangmycin on TRAIL-Induced Apoptosis in Gastric Cancer Cell Line AGS
【摘要】 目的探讨平阳霉素对TRAIL诱导的胃癌细胞株AGS凋亡的影响及机制。方法在培养的AGS细胞中加入100 ng·mL-1 TRAIL刺激,并分别加入20、40、80、160μg·mL-1的平阳霉素培养48 h,采用RT-PCR及Western blot检测survivin、bcl-2和bax mRNA及蛋白含量,TUNEL法检测AGS细胞凋亡,Western blot检测JNK蛋白及磷酸化水平。结果 40μg·mL-1平阳霉素可显著抑制AGS细胞survivin mRNA及蛋白表达(P<0.05),显著促进JNK1/2磷酸化水平升高(P<0.05),显著提高AGS细胞凋亡率(P<0.05)。80μg·mL-1平阳霉素可显著促进bax mRNA及蛋白表达(P<0.05)。结论平阳霉素可通过JNK信号通路促进TRAIL诱导的AGS细胞凋亡。
【Abstract】 Objective To investigate the effect of pingyangmycin(PYM) on TRAIL-induced apoptosis in gastric cancer cell line AGS.Methods AGS cells were treated with TRAIL(100 ng·mL-1) combined with PYM(20,40,80 or 160 μg·mL-1) for 48 hours.The mRNA and protein expression levels of survivin,Bcl-2 and Bax were determined by Western blot and RT-PCR,respectively.The apoptosis of AGS cells was measured by TUNEL assay.The expression levels of JNK and phosphorylated JNK were determined by Western blot.Results The treatment with 40 μg·mL-1 PYM suppressed survivin mRNA and protein expression,induced JNK1/2 phosphorylation and increased apoptosis in AGS cells(P<0.05).The treatment with 80 μg·mL-1 PYM up-regulated the expression of Bax mRNA and protein(P<0.05).Conclusion PYM treatment promotes TRAIL-induced apoptosis in AGS cells through activation of JNK pathway.
