筋骨草超微粉对LPS致急性肺损伤肉鸡的保护作用
Protective effects of ultramicro-pulverised powder of Ajuga decumbens on broilers with LPS-induced acute lung injury
【摘要】 将供试的200只1日龄白羽肉鸡随机分为5组(空白对照组、模型组、试验Ⅰ~Ⅲ组),每组5个重复,每个重复8只鸡.空白对照组、模型组饲喂基础日粮;试验Ⅰ、Ⅱ、Ⅲ组在饲喂的基础日粮中分别添加500、1 000、2 000 mg·kg-1筋骨草超微粉.试验为期42 d,研究筋骨草超微粉对脂多糖(LPS)引起的急性肺损伤肉鸡肺组织的保护作用.结果表明:注射LPS 0 h,试验Ⅰ~Ⅲ组肉鸡肺组织的IL-1β、IL-6、TNF-α含量较空白对照组显著提高(P<0.05);注射LPS 12、24 h,试验Ⅰ~Ⅲ组肉鸡血清的NO浓度、一氧化氮合酶(NOS)活性,肺组织的IL-1β含量、IL-6含量、TNF-α含量、髓过氧化物酶(MPO)活性及核蛋白质的NF-κBp65含量较模型组显著降低(P<0.05),肺组织chTLR4、NF-κB mRNA的表达量也显著下降(P<0.05);病理切片显示,模型组肉鸡肺组织的结构明显遭到破坏,试验Ⅰ~Ⅲ组较模型组有不同程度的改善.综上,筋骨草超微粉对LPS引起的急性肺损伤肉鸡的肺组织具有保护作用.在本试验条件下,筋骨草超微粉在肉鸡基础日粮中的适宜添加量为1 000 mg·kg-1.
【Abstract】 In order to explore the efficacy and mechanism of ultramicro-pulverised powder of Ajuga decumbens Thunb on broilers undergo acute lung injury induced by lipopolysaccharide(LPS), 100 1-day-old avine broilers were randomized into 5 groups, which included control group, model group, and experimental groups Ⅰ, Ⅱ and Ⅲ, with 5 replicates per group and 8 chickens per replicate. The broilers were fed a basal diet which was supplemented with 0, 0, 500, 1 000 and 2 000 mg·kg-1 A.decumbens ultramicro powder, respectively for 42 days. Acute lung injury models were established via intratracheal instillation of LPS. The results showed that IL-1β, IL-6 and TNF-α levels increased significantly in the lung of broilers in groups Ⅰ-Ⅲ once LPS was injected compared with the control(P<0.05). Twelve and 24 h after injection, serum NO level and nitric oxide synthase(NOS) activity, contents of IL-1β, IL-6, TNF-α, myeloperoxidase(MPO) activity, and NF-κBp65 in the lung went down significantly in groups Ⅰ-Ⅲ compared with the model group(P<0.05), so did the levels of chTLR4 mRNA and NF-κB mRNA(P<0.05). Pathological slides demonsrtacted that the lung tissues of broilers in the model group were damaged remarkably while the deterioration was mitigated in groups Ⅰ-Ⅲ. Therfore, ultramicro-pulverised powder of A.decumbens plays a protective role when healing broilers with LPS-induced acute lung injury. The recommended dosage is 1 000 mg·kg-1 A.decumbens for every kilogram of basal diet.
