防己黄芪汤中18种成分的UPLC-MS/MS定量分析

【摘要】 目的:建立超高效液相色谱-三重四级杆质谱联用(UPLC-QQQ-MS/MS)技术测定防己黄芪汤中18种成分含量的测定方法,为科学评价防己黄芪汤的质量提供依据。方法:采用UPLC-QQQ-MS/MS技术建立对防己黄芪汤中多成分同时测定的分析方法,对复方进行多成分表征。结果:建立了灵敏度高、分析质量可靠、分析效率高且能对防己黄芪汤复方中多成分同时测定的UPLC-QQQ-MS/MS分析方法,并应用于防己黄芪汤中的生物碱类(木兰花碱、防己诺林碱、粉防己碱)、皂苷类(黄芪皂苷Ⅲ、黄芪甲苷、黄芪皂苷Ⅰ、甘草次酸)、黄酮类(毛蕊异黄酮葡萄糖苷、芒柄花苷、5-O-甲基维斯阿米醇苷、毛蕊异黄酮、芒柄花素、甘草苷、异甘草苷、甘草素、异甘草素)和萜类(白术内酯Ⅲ、白术内酯Ⅱ)共18种成分的定量分析研究,其中粉防己碱为防己黄芪汤复方中含量最高的生物碱。结论:该方法专属性强,可用于防己黄芪汤中多种成分的定量检测。

【Abstract】 Objective: To establish a method for determining eighteen components in Fangji Huangqi Decoction(FJHQD) by UPLCQQQ-MS/MS, in order to provide a basis for the scientific evaluation of the quality of FJHQD. Methods: A method for simultaneous determination of multi-components in FJHQD by UPLC-QQQ-MS/MS technology was established to characterize the multi-components of the compound. Results: A UPLC-QQQ-MS/MS analysis method for simultaneous determination of multi-components in FJHQD, with high sensitivity, reliable analysis quality, and high efficiency, was established. This method was applied to study the quantitative analysis of 18components in FJHQD, including alkaloids(magnoflorine, fangchinoline, tetrandrine), saponins(astragaloside Ⅲ, astragaloside Ⅳ, astragalosideⅠ, glycyrrhetic acid), flavonoids(calycosin-7-glucoside, ononin, 5-O-methylvisammioside, calycosin, formononetin, liquiritin, liquiritigenin, neoisoliquritin, isoliquiritigenin), and terpenes(atractylenolide Ⅲ, atractylenolide Ⅱ). Additionally, tetrandrine was the alkaloid with the highest content in FJHQD compound. Conclusion: This method is specific and can be used for the quantitative determination of various components in FJHQD.
