羧甲基聚合-β-环糊精作为选择剂应用于毛细管电泳法拆分手性化合物
Use of Carboxymethyl-poly-β-cyclodextrin as Chiral Selective Agent in Capillary Electrophoretic Separation of Chiral Compounds
【摘要】 提出了一种用毛细管电泳拆分3种未衍生化的氨基酸对映体(苯丙氨酸,酪氨酸,色氨酸)和一种手性药物(芬氟拉明)的方法。以水溶性羧甲基聚合-β-环糊精为手性选择剂,采用毛细管区带电泳模式,考察了手性选择剂浓度、缓冲溶液pH值、柱温及电压对分离的影响。4种手性化合物在各自优化的试验条件下,均达到基线分离。此法操作简单,可用于这4种手性化合物的质量控制。
【Abstract】 Capillary electrophoresis(CE) was applied to enantiomorphous separation of 3 un-derivatized amino acids,i.e.,phenylalanine,tyrosin,tryptophan,and a chiral drug,"Fenfluramine",and the water soluble carboxymethyl-poly-β-cyclodextrin(CM-poly-β-CD) was used as chiral selective agent.Under the mode of zone electrophoresis,conditions for the separation,including the concentration of CM-poly-β-CD,pH-value of buffer solution,temperature and pressure of the column were tested individually and optimized.Under the respective optimum condition,enantiomorphous separation of each of the 4 chiral compounds was attained and clear separation from the baseline was observed.The proposed method was found feasible to be applied to quality control of the 4 chiral compounds.
