基于网络药理学的如意金黄散对坏死性筋膜炎的治疗机制研究
Study on the therapeutic mechanism of Ruyi Jinhuang Powder on necrotizing fasciitis based on network pharmacology
【摘要】 目的 拟使用网络药理学工具研究如意金黄散对坏死性筋膜炎的作用机制。方法 在TCMSP和DrugBank等数据库中收集如意金黄散可能的药效成分和作用靶点以及坏死性筋膜炎的作用靶点并取交集,使用Cytoscape软件构建“草药-成分-靶点”互作网络。在STRING数据库构建蛋白互作网络并筛选出如意金黄散核心作用靶点,使用GO和KEGG对如意金黄散治疗坏死性筋膜炎的作用机制进行分析,使用分子对接验证预测药效成分与核心靶点的亲和能力。结果 如意金黄散可通过44个靶点对坏死性筋膜炎进行治疗,其治疗的核心靶点主要为ALB、IL-6、TNF、INS、STAT3和PTGS2,可通过IL-17、TNF、NF-κB等信号通路抑制急性炎症反应、细胞因子调节及内毒素反应,且药效成分与预测靶点具有良好的亲和力。结论 如意金黄散可通过“多靶点-多途径”方式治疗坏死性筋膜炎,为如意金黄散治疗坏死性筋膜炎的临床应用提供了理论依据。
【Abstract】 Objective To study the mechanism of action of Ruyi Jinhuang Powder on necrotizing fasciitis by network pharmacology. Methods The potential components and possible targets of Ruyi Jinhuang Powder and necrotizing fasciitis were collected from TCMSP and DrugBank databases, and their intersection was selected. The “herb-component-target” interaction network was constructed using Cytoscape software. The protein interaction networks were constructed in STRING database, and core targets of Ruyi Jinhuang Powder were screened. GO and KEGG were used to analyze the mechanism of action of Ruyi Jinhuang Powder in the treatment of necrotizing fasciitis, and molecular docking was used to validate the ability to predict affinity between components and core targets. Results Ruyi Jinhuang Powder could treat necrotizing fasciitis through 44 targets, the core targets of which were mainly ALB, IL-6, TNF, INS, STAT3, and PTGS2. It could inhibit acute inflammatory response, cytokine regulation, and endotoxin response through IL-17, TNF, NF-κB and other signaling pathways, and the active ingredients of Ruyi Jinhuang Powder had good affinity with predicted targets. Conclusion Ruyi Jinhuang Powder can treat necrotizing fasciitis through “multi-target-multi-pathway” approach. This study provides a theoretical basis for the clinical application of Ruyi Jinhuang Powder in the treatment of necrotizing fasciitis.
