中药小分子介导铁死亡在消化道肿瘤中的作用机制研究进展
Research Progress on Mechanism of Ferroptosis Mediated by Small Molecules of Traditional Chinese Medicine in Gastrointestinal Tumors
【摘要】 铁死亡是当今医学领域研究热点,是一种由铁和脂质过氧化驱动的新型细胞死亡模式,受铁代谢、氨基酸代谢、脂代谢等多种细胞代谢途径的调节。越来越多的证据表明,通过调控铁死亡通路中的相关靶点和通路能够抑制肿瘤生长,增强消化道抗肿瘤对放化疗的敏感性,降低消化道肿瘤细胞的耐药性。我国是天然的中草药宝库、种类繁多。研究发现多成分、多靶点、毒性小、药理作用广的中药小分子能够促进铁死亡发生,如青蒿素及衍生物、萜类、黄酮类、生物碱类等,直接或间接地诱导铁死亡的发生,从而抑制相关癌细胞的生长和增殖,在抗消化道肿瘤发挥重要作用。从中药小分子通过调控铁死亡在消化道抗肿瘤作用机制研究中的进展进行综述,以期为中医药治疗消化道肿瘤提供新的理论依据和研究思路。
【Abstract】 Ferroptosis is a new type of cell death driven by iron and lipid peroxidation, which is regulated by a variety of cell metabolic pathways such as iron metabolism, amino acid metabolism and lipid metabolism. More and more evidence shows that regulating the related targets and pathways in the ferroptosis pathway can inhibit tumor growth, enhance the sensitivity of gastrointestinal anti-tumor to radiotherapy and chemotherapy, and reduce the drug resistance of gastrointestinal tumor cells. China is a natural treasure trove of Chinese herbal medicine with a wide variety of species. Studies have found that small Chinese medicine molecules with multiple components, multiple targets, low toxicity and wide pharmacological effects can promote iron death, such as artemisinin and its derivatives, terpenoids, flavonoids and alkaloids, directly or indirectly induce iron death, thereby inhibiting the growth and proliferation of related cancer cells, and playing an important role in anti-gastrointestinal tumors. This article reviewed the research progress of the anti-tumor mechanism of small molecules of traditional Chinese medicine by regulating ferroptosis in gastrointestinal tumors, so as to provide a new theoretical basis and research ideas for the treatment of gastrointestinal tumors with traditional Chinese medicine.
