小麦α-淀粉酶抑制剂降血糖作用的实验研究

【摘要】 目的:研究从小麦中提取的α-淀粉酶抑制剂对糖尿病小鼠的影响。方法:四氧嘧啶性糖尿病小鼠灌胃给予α-淀粉酶抑制剂45 mg·kg-1,测定其淀粉耐量和糖耐量;四氧嘧啶性糖尿病小鼠灌胃给予α-淀粉酶抑制剂(5,15,45mg·kg-1),连续21 d后,测定血糖值和小肠内二糖酶活性。血清葡萄糖测定采用葡萄糖氧化酶法。结果:α-淀粉酶抑制剂能够增加四氧嘧啶性糖尿病小鼠的淀粉耐量而对其葡萄糖耐量没有影响;糖尿病小鼠灌服α-淀粉酶抑制剂45和15 mg·kg-1后,血糖值均明显低于模型组。α-淀粉酶抑制剂能有效抑制小鼠小肠内各肠段的麦芽糖酶和蔗糖酶活性。结论:小麦α-淀粉酶抑制剂对糖尿病小鼠有降血糖作用,其机制可能与抑制小肠内各肠段的麦芽糖酶和蔗糖酶活性有关。

【Abstract】 Objective:To assess the anti-hyperglycemic effect of a-amylase inhibitor (AI) isolated from wheat in diabetic mice. Methods:Diabetic mice administered with amylase inhibitor (5 , 15 and 45 mg·kg-1) by gastric perfusion for 21 days were monitored the blood glucose level and disaccharidase activity. Diabetic mice treated with AI 45 mg·kg-1 by gastric perfusion were tested with the starch and sugar tolerance. Blood glucose level was measured using glucose oxidase assay. Results: AI showed an increase on the starch tolerance but no effect on the sugar tolerance. AI (15 and 45 mg·kg-1) significant- ly reduced blood glucose level of diabetic rats and significantly inhibited the disaccharidase activity in various segments of mice small intestine. Conclusion: AI decreases the blood glucose level of diabetic mice by inhibiting the disaccharidase activity in mice small intestine.
