迷迭香精油纳米乳的制备研究

【摘要】 目的:制备迷迭香精油(Rosemary essential oil,REO)纳米乳,增强其稳定性并增加其抑菌活性。方法:采用气相色谱-质谱联用技术(GC-MS)分析REO成分,通过拟三元相图确定REO纳米乳的最佳制备工艺,以粒径和PDI为指标对其表征,采用琼脂扩散法测定其抑菌活性。结果:GC-MS分析共鉴定出45种成分,主要为烯醇类化合物;经筛选得出REO纳米乳的最佳制备工艺为:15.50%的聚氧乙烯蓖麻油,15.50%的无水乙醇,6.90%的REO以及62.10%的蒸馏水;REO纳米乳平均粒径为18.43±0.50nm,PDI为0.09±0.03;抑菌实验表明REO及REO纳米乳对金黄色葡萄球菌、大肠杆菌、白色念珠菌有显著抑制作用(p<0.05)。结论:本研究成功制备REO纳米乳,稳定性好,有较好的推广前景,为REO的推广和应用提供依据。

【Abstract】 Objective:Rosemary essential oil(REO)nanoemulsion was prepared to enhance its stability and increase its antibacterial activity.Methods:The components of REO were analyzed by gas chromatography-mass spectrometry(GC-MS).The optimum preparation process of REO nano emulsion was determined by pseudo ternary phase diagram.It was characterized by particle size and PDI,and its antibacterial activity was determined by agar diffusion method.Results:45 components were identified by GC-MS,mainly olefins and alcohols;The optimum preparation process of REO nano emulsion was 15.50% polyoxyethylene castor oil,15.50% absolute ethanol,6.90% REO and 62.10% distilled water;The average particle size of REO nanoemulsion was 18.43±0.50nm and PDI was 0.09±0.03;Bacteriostatic experiments showed that REO and REO nano emulsion had significant inhibitory effects on Staphylococcus aureus,Escherichia coli and Candida albicans(p<0.05).Conclusion:REO nanoemulsion was successfully prepared in this study.Compared with essential oil,REO nanoemulsion had good stability.It has a good promotion prospect,which provides a basis for the promotion and application of REO.
