电晕电场将大豆Gy3基因导入花棒愈伤组织及植株再生
Introduction of Soybean Gy3 Gene into Callus of Hedysarum scoparium by Corona Discharge and Plantlet Regeneration
【摘要】 以花棒幼胚诱导的愈伤组织为受体材料,利用针-板电晕电场将pBI121质粒(含GUS基因和卡那霉素抗性基因)导入花棒愈伤组织,利用组织化学定位实验检测到GUS基因在受体组织中的瞬时表达。用80 mg·L-1卡那霉素筛选转基因花棒愈伤组织中含有的大豆Gy3基因,获得了38株再生植株,含大豆Gy3基因3株,再生植株经PCR、RT-PCR检测初步证实大豆Gy3基因已转入花棒植物基因组DNA中并发生转录。
【Abstract】 The pBI121 plasmid,which contains a copy of GUS gene and kanamycin resistance gene,was introduced into Hedysarum scoparium calli by corona discharge,and transient expression of GUS gene was detected and discovered in these receptor calli by histochemical localization reaction.The soybean Gy3 gene in Hedysarum scoparium transformation calli was screened by Kanamycin of 80 mg·L-1,and 38 regenerated plants were obtained,and there were soybean Gy3 gene in 3 plants.PCR and RT-PCR detection showed that the Soybean Gy3 gene had been introduced into DNA of the regenerated plants of Hedysarum scoparium successfully.
