GATA结合蛋白3、性别决定相关基因簇2在三阴性乳腺癌中的表达及预后关系

【摘要】 目的 探讨GATA结合蛋白3(GATA3)、性别决定相关基因簇2(SOX2)在三阴性乳腺癌(triple-negative breast cancer,TNBC)中表达及预后关系。方法 收集新疆肿瘤医院140例TNBC和癌旁组织标本,通过免疫组化法染色检测GATA3和SOX2在TNBC中的表达水平,分析GATA3及SOX2在TNBC中的表达特点、临床病理特征及预后的关系。结果 GATA3在TNBC及癌旁组织中的表达率为(82.86%、98.00%),SOX2在TNBC及癌旁组织中的表达率分别为(56.43%、5.00%),二者差异均有统计学意义(P<0.05)。GATA3阳性表达与TNBC患者低组织学分级及淋巴结转移有关(P<0.05),SOX2阳性表达与TNBC患者出现低组织学分级、Ki-67增殖表达具有明显相关性(P<0.05)。GATA3阳性表达及SOX2阳性表达者5年无病生存率分别为78.35%(91/116)、62.02%(49/79),差异有统计学意义(P<0.05)。结论 TNBC中GATA3阳性率明显低于癌旁组织,SOX2阳性率高于癌旁组织。GATA3缺失表达及SOX2过表达是乳腺癌患者预后差的免疫组化指标,有望成为乳腺癌患者预后评估的重要依据。

【Abstract】 Objective To investigate the expression and prognostic relationship of GATA binding protein 3(GATA3)and sex determination related gene cluster 2( SOX2) in triple-negative breast cancer( triple-negative breast cancer,TNBC). Methods A total of 140 cases of TNBC and adjacent cancer tissue were collected, and the expression levels of GATA3 and SOX2 in TNBC were detected by immunohistochemical staining, and the characteristics of GATA3 and SOX2expression, clinicopathological characteristics and prognosis in TNBC were analyzed. Results The expression rate of GATA3 in TNBC and adjacent tissues(82. 86%, 98. 00%), and the expression rate of SOX2 in TNBC and adjacent tissues were( 56. 43% and 5. 00%, respectively), which were statistically significant(P< 0. 05). Positive GATA3expression was associated with low histological grade and lymph node metastasis in TNBC patients(P<0. 05), and positive SOX2 expression showed low histological grade and Ki-67 proliferative expression in TNBC patients(P<0. 05). The 5-year disease-free survival rate of patients with GATA3 positive expression and SOX2 positive expression was 78. 35%(91/116)and 62. 02%(49/79), respectively, which were statistically significant(P<0. 05). Conclusion GATA3 positive rate in TNBC is significantly lower than in adjacent tissues, and SOX2 positive rate is higher than adjacent tissues. GATA3deficiency expression and SOX2 overexpression are immunohistochemical indicators of poor prognosis of breast cancer patients, and are expected to be an important basis for prognosis evaluation of breast cancer patients.
